摘 要:目的:观察心脑宁片对 D-半乳糖致小鼠亚急性衰老模型胸腺组织形态的影响。方法:取体质量18~21g小鼠72只,雌雄各半,其中 12只为空白对照组,余 60只每天颈背部皮下注射 D半乳糖(生理盐水配成质量浓度为 5%,注射量为 0.025mL·g^-1)制作糖代谢衰老模型,每天给药 1次,连续 40d。模型制作成功后,随机平均分为模型组 (生理盐水0.02mL·g^-1),脑安片组(0.53g·kg^-1,26.5g·L^-1,0.02mL·g^-1,相当于临床用量的15倍),大剂量、中剂量、小剂量心脑宁片组(2.24g·kg^-1、1.12g·kg^-1、0.56g·kg-1,112g·L^-1、56g·L^-1、28g·L^-1,0.02mL·g^-1,相当于临床剂量的 30倍、15倍、7.5倍)。每天给药1次,连续30d。取小鼠胸腺组织用福尔马林固定,石蜡包埋,切片,HE染色,光镜观察胸腺组织形态的变化。结果:模型组胸腺皮质厚度、淋巴细胞数分别为(7.26±2.18)μm、(21.4±3.7)个,脑安片组分别为(23.18±945)μm、(36.3±12.2)个,大剂量心脑宁片组分别为(27.89±3.46)μm、(46.6±10.4)个,中剂量心脑宁片组分别为(2436±419)μm、(37.5±8.60)个,小剂量心脑宁片组分别为(22.18±3.45)μm、(36.3±7.7)个,空白对照组分别为(2862±435)μm、(57.3±11.4)个。各组胸腺皮质厚度、淋巴细胞数与模型组比较,差异均有统计学意义(P<0.01)。结论:心脑宁片可显著增加 D半乳糖致小鼠亚急�Objective:To observe the effect of Xinnaoning Tablet on thymus tissue of D-galactose induced subacute aging model. Methods :72 mice, male and female half, 12 for blank control group, the rest 60 mice were given back neck subcutaneous injection of D-galactose( with physiological saline into mass concentration 5% , injection volume of 0. 025 mL · g^- 1 ) to form aging model, 1 times a day ,40 D in a row. After the success of the model making, randomly divided into model group ( physiological saline 0.02mL · g^- 1 ) , Naoan group (0.53 g · kg^-1,26.5g · kg^-1,0.02 mL · g^-1, 15 times of clinical dosage),large dose group, middle dose and small dose of Xinshenning (2.24 g · kg^-1 ,1.12 g · kg^-1 ,0.56 g · kg^-1 ,112 g · kg^-1 ,56g · kg^-1 ,28 g · kg^-1,0.02 mL · g^- 1,equivalent to 30 times, 15 times ,7.5 times of clinical dosage) , once a day for 30 d. Thymus tissue in mice with formalin fixed, paraffin embedding, section, HE stain, light microscopy to observe the organization change. Results : The thymus cortex thickness and lymphocyte count of modelgroup respectively was (7.26 ± 2.18 ) um and (21.4 ± 3.7 ), in Naoan group respectively was ( 23.18 ± 9.45 ) um and ( 36.3 ± 12. 2 ) , in large dose group of Xinnaoning was respectively ( 27.89 ± 3.46 )um and ( 46.6 ± 10.4 ) , in middle dose group was respectively (24.36 ± 4.19 )um and (37.5 ± 8.60) ,in small dose group was respectively (22.18 ± 3.45 ) um and (36.3 ± 7.7 ) and in blank control group was respectively ( 28.62 ± 4.35 ) um and ( 57.3 ± 11.4 ). Compared with model group, the difference had statistical significance (P 〈 0.01 ). Conclusion:Xinnaoning Tablet can significantly increase the thymus tissue thickness and lymphoeytes number of D-galactose induced subacute aging model mice with good anti-aging effect.
