摘  要:目的:采用响应曲面法优化毛果鱼藤中香豆素类化合物的提取工艺,提高香豆素类化合物的提取与分离效率。方法:以毛果鱼藤藤茎为原料,采用Box-Behnken设计实验,结合微波提取法,分别从料液比、乙醇体积分数和提取时间3个方面考察其对香豆素类化合物提取效果的影响,以优化提取工艺;以毛果鱼藤中香豆素衍生物Robustin acid为考察指标,采用高效液相色谱法进行检测。结果:毛果鱼藤中香豆素类化合物的优化提取工艺条件为微波功率为400 W,提取温度为70℃,提取时间为11.67min,料液比为1∶49.67,乙醇体积分数为86.67%,香豆素提取率为8.90 mg·g-1;高效液相的最佳色谱条件为:检测波长为254nm,柱温为30℃,流动相为乙腈-0.5%醋酸水溶液,梯度洗脱分离效果最佳。结论:以毛果鱼藤中香豆素衍生物Robustin acid高效液相色谱为参考,采用Box-Behnken响应曲面法优化了香豆素类化合物的提取工艺,提高了此类化合物的提取效率,为分离毛果鱼藤藤茎中更丰富的香豆素类化合物奠定了基础。Objective: To optimize extraction process for coumarins from the stem of Derris eriocarpa by response surface methodology,and improve extraction efficiency and productivity of coumarins from the stem of Derris eriocarpa. Methods: Extraction process of coumarins from the stem of Derris eriocarpa was optimized by Box- Behnken response surface method combined with microwave extraction. Three aspects of ethanol volume fraction,solid- liquid ratio and extraction time were investigated to evaluate their influence on extraction. The detection was performed by HPLC with the coumarin derivative Robustin acid as the investigation index. Results: The optimum conditions for extraction of coumarins were found to be 400 W,70 ℃,11. 67 min,1∶ 49. 67( g·m L- 1),and 86. 67% for extraction power,extraction temperature,extraction duration,solid- liquid ratio,and volume fraction of ethanol,respectively.The efficiency of extraction under these optimum conditions was found to be 8. 90 mg·g- 1of coumarins. The optimum HPLC conditions were the mobile phase of acetonitrile- 0. 5% acetic acid,the temperature of 30 ℃ and the UV detection wavelength of 254 nm to achieve the best separation by gradient elution. Conclusion: The optimized processing parameters of coumarin compounds were acquired by the Box- Behnken response surface method with reference to the HPLC profile of the coumarin deriative Robustin acid. This optimal experiment provided the optimal conditions to improve efficiency and productivity of coumarins and laid a foundation for separation of rich varieties of coumarin compounds in the stem of Derris eriocarpa How.
