异叶青兰提取物抗病毒作用及挥发油成分分析

摘　　要：目的:探讨藏药异叶青兰挥发油、石油醚提取物、乙酸乙酯提取物、正丁醇提取物、水提取物抗腺病毒(ADV)、流感甲型病毒A/汉防/359/95(H3N2)及单纯疱疹病毒II型(HSV-2)的作用。方法:细胞病变法测定提取物的半数有毒浓度及对病毒的半数效应浓度,以治疗系数(TI)为指标评价药效。采用气相色谱-质谱联用分析挥发油成分。结果:石油醚提取物抗A/汉防/359/95(TI=12.99)、抗单纯疱疹病毒II(TI=6.48)活性最强。挥发油抗腺病毒(TI=7.77)活性最强。挥发油对单纯疱疹病毒II型有较强的直接灭活作用(TI=3.1)。挥发油主要成分为桉油精(22.18%),桃金娘烯醇(11.28%),顺式松油醇(11.11%)等。结论:异叶青兰挥发油、石油醚提取物、乙酸乙酯提取物、正丁醇提取物、水提取物具有一定的抗腺病毒、流感甲型病毒A/汉防/359/95(H3N2)及单纯疱疹病毒II型的作用。Objective： To analyze components in the essential oil and various extracts from Tibetan herbal medicine Dracocephalum heterophyllum Benth, and evaluate their antiviral effects. Methods： The essential oil, nbutanol extract, ethyl acetate extract, aqueous extract and petroleum ether extract were obtained from Dracoceph- alum heterophyllum Benth. Their effects on adenovirus A/hanfang/359/95 （H3N2） and herpes simplex virus type 2 were evaluated. The cytotoxicity and the antiviral effectiveness were investigated using cytopathic effect （CPE）o The therapeutic index （TI） was used to assess antiviral activity. At the same time, the components of volatile oil were analyzed by gas chromatography-mass spectrometry. Results： The petroleum ether extract had the strongest activities against A/hanfang/359/95 （ H3N2） （TI = 12.99） and herpes simplex virus type 2 （TI = 6.48 ）. The essential oil had the strongest activity against adenovirus （TI = 7.77 ）, and a direct killing activity against herpes sireplex virus type 2 （TI = 3.1 ） suggesting it m t be a strong direct inactivator. The essential oil contained eucalyptol （22.18%）, 6,6-dimethyl-bicyclo [3.1.1 ] hept-2-ene-2-methanol （ 11.28% ）, cis-terpineol （ 11.11% ）, and so on. Conclusion： The essential oil, n-butanol extract, ethyl acetate extract, aqueous extract and petroleum ether extract from Tibetan herbal medicine Dracocephalum heterophyllum Benth have antiviral effects against adenovirus, A/hanfang/359/95 （H3N2） and herpes simplex virus type 2 in vitro.
