摘 要:目的:用两相溶剂萃取法从金雀根提取物中纯化二苯乙烯有效部位,对其工艺条件及参数进行考察,优选出最佳纯化工艺。方法:以二苯乙烯有效成分carasinol B,kobophenol A,(+)-α-viniferin在萃取物中的含量及萃取保留率为指标,对萃取原液的浓度、乙酸乙酯用量、萃取次数、萃取时间进行考察。通过单因素考察结合正交试验优选最佳纯化工艺。结果:最佳纯化工艺为:萃取原液浓度为0.25 g·m L-1,乙酸乙酯用量为1倍原液量,萃取2次,每次20 min。得到的3批萃取物有效成分含量平均为69.18%,保留率为85.21%。结论:该纯化工艺合理可行,可用于金雀根中二苯乙烯有效部位的纯化。Objective: To determine and optimize the process conditions of separating and purifying stilbene effective parts from Genistein root using two-phase solvent extraction. Methods: The concentration of extraction solution, extraction times, amount of extraction, and extraction time were studied, and the percentage of active ingredients in the extraction and extraction ratio were used as indexes. The concentration of stilbene active ingredients (carasinol B, kobophenol A, ( + )-α-viniferin) was determined by HPLC, and the optimized purification parameters were obtained by single factor and orthogonal experiments. Results: The optimized purification conditions were as follows : extraction solution concentration was 0.25 g·mL^-1, one fold of ethyl acetate was used, extraction was done for two times and 20 min for each time. The average percentage of active ingredients in extraction of three batches was 69.18% , and extraction ratio was 85.12%. Conclusion: The purification process is reasonable and practicable, and can be used for purification of stilbene active ingredients from Genistein root.
