关中黑猪精液常温保存研究

摘　　要：关中黑猪是陕西省培育品种,具有抗病能力强、繁殖力高、耐粗饲和肉质好等特点。由于国外瘦肉型猪对国内市场的冲击,关中黑猪的市场环境日益下降,使得许多优良性状发生分离变化。本研究选取5种常用猪精液常温保存稀释剂,对关中黑猪精液保存最适宜温度及抗生素选择进行研究。结果表明,关中黑猪种公猪鲜精活力为0.85,畸形率16%,pH7.3,精子密度2.36亿/mL,平均采精量250mL,色泽为乳白色。在不同的稀释剂中,保存效果最好的是Ⅲ号稀释剂,可保存5d,活力为0.5,Ⅰ号稀释剂效果次之,Ⅱ号稀释液略好于Ⅳ、Ⅴ号稀释液。在短时间内（≦2d）,Ⅰ号稀释剂与Ⅲ号稀释剂保存效果无明显差异,而Ⅰ号稀释剂成本更低,更适用于生产实践中关中黑猪精液短期保存。此外,添加青霉素、庆大霉素或青霉素＋庆大霉素的混合液后保存效果没有差异。因此,关中黑猪精常温保存的最佳温度是15~20℃,本研究为关中黑猪人工授精技术及提高育种水平提供依据。Guanzhong Black pig is Shaanxi province cultivars, and an excellent dual-purpose breed of meat and fat, with strong disease resistance, high fecundity, resistance to crude feed and good meat quality. In recent decades, due to the impact of foreign pigs on the domestic market, the market environment of Guanzhong Black pig declined, which makes many good traits separating occur. The test for this situation, the boar semen quality testing and stored at room temperature study of Guanzhong Black pig provides a theoretical basis for artificial insemination and the improvement of breeding level. In order to select the greatest formula of diluents, in the present study, 5 kinds of diluents of semen preservation in normal temperature were selected to compare the effect of preservation with each other. And we also investigated differences of different antibiotics and the best preservation temperature for Guanzhong Black pig semen on the basis of the greatest formula. In this study, we collected the semen of Guanzhong Black stock boar by an artificial vagina, and then detected it, the motility of sperm was 0.85 ; the deformity rate was 16V00~ pH is 7.3; and density was 0. 236 billion per milliliter; the volume of semen was 250 mL; color was milkiness. Screened all dilutions for preservation results, we found that the dilution III was the most effective, dilution I was the second one, dilution II was slightly better than dilution IV and dilution V. The effect of preservation between dilution I and dilution III was not significant in a short time （42 d）, moreover, the cost of dilution I which more suitable for the pro- duct/on practice was lower. In addition, the best preservation temperature of Guanzhong Black pig semen was 15~20℃, and there was no different in preservation after adding penicillin or gentamicin or the mixture of penicillin and gentamicin.
