一贯煎对大鼠肝纤维化拮抗作用的影响

摘　　要：目的:探讨一贯煎对大鼠肝纤维化的拮抗作用。方法:将SD大鼠随机分为正常组、模型组、秋水仙碱组、一贯煎组。除正常组外,其余3组均建立四氯化碳(CCl4)诱导的肝纤维化模型。造模后,一贯煎组和秋水仙碱组予相应药物灌胃、正常组和模型组以0.9%氯化钠溶液灌胃,灌胃次数为1次/d。灌胃4周后,检测大鼠肝组织基质金属蛋白酶-1(MMP-1)、金属蛋白酶抑制因子-1(TIMP-1),通过病理染色评价肝纤维化程度。结果:与正常组相比,模型组肝纤维化程度增加,MMP-1降低、TIMP-1升高、MMP-1/TIMP降低(P<0.05,P<0.01)。与模型组相比,一贯煎组及秋水仙碱组肝纤维化程度减轻,TIMP-1降低(P<0.05);秋水仙碱组MMP-1有降低趋势,一贯煎组MMP-1有升高趋势;两治疗组MMP-1/TIMP-1有升高趋势。结论:一贯煎可拮抗CCl4诱导的大鼠肝纤维化,其机制可能与调节肝组织内MMP-1/TIMP-1含量,促进胶原纤维降解有关。Objective： To investigate the antagonistic effect of Yiguanjian Decoction on hepatic fibrosis in rats. Methods： SD rats were randomly divided into normal group, model group, colchicine group and Yiguanjian Decoction group. Except for the normal group, the other three groups were established as the model of hepatic fibrosis induced by carbon tetrachloride （CC14）. After modeling, Yiguanjian Decoction group and colchicine group were garaged by corresponding drug, while the normal group and model group were treated with normal saline, once a day. After 4 weeks of treatment, the level of matrix metalloproteinase-1 （MMP-1） and tissue inhibitors of metalloproteinase-1 （TIMP-1） in liver tissue were detected, and the liver fibrosis degree was evaluated by pathological staining. Results： Compared with the normal group, the liver fibrosis degree in model group was increased, the level of MMP-I was decreased, the level of TIMP-1 was increased and the ratio of MMP-1 to TIMP-1 was decreased （P〈0.05, P〈0.01）. Compared with the model group, the liver fibrosis degree in colchicine group and Yiguanjian Decoction group were both decreased, the level of TIMP-1 was decreased （P〈0.05）, the level of MMP-1 in colchicine group was decreased and in Yiguanjian Decoction group was increased. The ratio of MMP-1 to TIMP-1 were both increased. Conclusion： Yiguanjian Decoction can antagonize the liver fibrosis in rats induced by CC14, which may be related to its regulating effect on MMP-1/TIMP-1 in liver tissue.
