摘 要:目的:探讨微波辐射对小鼠生精细胞膜流动性的影响及金匮肾气丸治疗机制。方法:雄性BALB/c小鼠50只,随机分为5组,每组10只。平行对照组不做任何处理;辐射对照组、金匮肾气丸1组、金匮肾气丸2组、金匮肾气丸3组均暴露于频率为2 450MHz连续微波中,功率密度为10m W/cm2,辐射18d,每天辐射1h;辐射对照组用0.9%氯化钠溶液分别灌胃,每天1次,每次0.1m L;金匮肾气丸组用金匮肾气丸(0.1、0.2、0.4g/kg体质量)分别灌胃,每天1次。组合酶法分离各组小鼠生殖细胞,荧光偏振法及激光扫描共聚焦显微镜(LSCM)检测膜微黏度(η)、荧光偏振度(P)。结果:微波辐射后,P及η显著升高(P<0.05);金匮肾气丸治疗后,P及η皆有显著改善(P<0.05),金匮肾气丸1组、2组、3组均接近正常,3组之间无显著差异。结论:微波辐射可以使小鼠生精细胞膜流动性下降,而金匮肾气丸的治疗机制是通过增加小鼠精子膜的流动性而起到治疗作用。Objective: To study the effects of microwave radiation on membrane fluidity of spermatogenic cells of male mice and the treatment mechanism of Jinkui Shenqi Pill. Methods: 50 male BALB/c mice were randomly divided into 5 groups, with 10 rlfice in each group. Except for the parallel control group, radiation control group, Jinkui Shenqi Pills group 1, Jinkoi Shenqi Pills group 2 and Jinkui Shenqi Pills group 3 were exposed to the continuous microwave of the frequency of 2 450 MHZ and the power density of 10mW/cm2 for one hour every day, lasting for 18 days. The mice in radiation control group were garaged with normal saline water 0.1mL once a day. Jinkui Shenqi Pills groups were gavaged with Jinkui Shenqi Pill once a day, group 1 with 0.1 gram per kilogram of body weight every time, group 2 with 0.2 gram per kilogram of body weight every time and group 3 with 0.4 gram per kilogram of body weight every time. The complex enzymatic method was used to separate the germ cells of mice in each group. The fluorescence polarization method and laser scanning confocal microscope (LSCM) were used to test membrane micro viscosity (η) and fluorescence polarization (P). Results: The membrane fluorescence polarization (P) and the membrane micro viscosity (η) after microwave radiation were significant higher than those before microwave radiation (P〈0,05). Compared with before treatment, the membrane fluorescence polarization (P) and the membrane micro viscosity (η) after Ueatment with Jinkui Shenqi Pill were improved significantly (P〈0.05). The mice in Jinkui Shenqi Pill group 1, 2 and 3 recovered, with no significant difference among the three groups. Conclusion: Microwave radiation can make decline membrane fluidity of spermatogenic cells in mice. The treatment mechanism of Jinkui Shenqi pills can increase the membrane fluidity of mice sperm.
