摘  要:目的:探讨当归散的不同化学组分对低子宫容受性小鼠受孕率的影响差异,筛选改善控制性超促排卵(COH)所致受孕率降低的最佳化学组分配方。方法:采用溶剂法对当归散中化学组分进行系统分离,分离出4个不同极性化学成分群,即石油醚提取物(组分A)、乙酸乙酯提取物(组分B)、乙醇提取物(组分C)和水提取物(组分D),并采用正交设计法配制出化学组分配方。采用"曲普瑞林+尿促性素(HMG)+绒促性素(HCG)"长周期超促排卵法制备小鼠低子宫容受性模型,造模同时预防性给予各化学组分配方,共9d。排卵当日合笼,于孕8d剖取子宫,清点胚泡。结果:当归散中组分A、B、D对受孕率具显著性影响,组分B、D使用可提高小鼠受孕率,而组分A则使受孕率降低。结论:当归散可以改善促排卵引起的子宫容受性降低,其中BCD化学组分配方为最优配方。Objective: Explorement of the effects of different chemical composition of Danggui Powder (DGS) on pregnancy rate in mice with low endometrial receptivity, to screen the optimum chemical composition which can improve the low pregnancy rate induced by COH. Methods: The solvent method was used to systematically extract the chemical composition of DGS. There were 4 groups of different polar chemical composition extracted from DGS, such as petroleum ether extract (component A), ethyl acetate extract (component B), ethanol extract (component C) and water extract (component D). The orthogonal design method was used to prepare the formulae comprised of different chemical composition. Mice were fed with Triptorelin+HMG+HCG, the method of ovulation induction for a long period, to establish the low endometrial receptivity mice model. At the same time, each group received the formulae comprised of different chemical composition, lasting for 9 days. The mice were mated during ovulation. After 8 weeks of pregnancy, the number of blastocysts in uterus was examined. Results: The component A, B and D of DGS had significant effects on the pregnancy rate. Component B and D increased mice pregnancy rate, but component A showed the opposite effect. Conclusion: Danggui Powder can improve the low endometrial receptivity induced by ovulation induction. And, the formula composed of component B, C and D may be the optimum chemical composition preparation.
