槐耳清膏增加人胃腺癌细胞MKN45对顺铂敏感性研究

摘　　要：目的:体外观察槐耳清膏在增加人胃腺癌MKN45细胞对顺铂(DDP)敏感性中的作用并探讨可能的机制。方法:MTT法观察不同浓度槐耳清膏、DDP单独或联合应用对MKN45细胞生长的抑制作用,并通过计算Q值判定两药合用的效果;用流式细胞仪检测槐耳清膏与DDP联用对MKN45细胞凋亡和细胞周期的影响。结果:单药槐耳清膏及DDP均能抑制MKN45细胞的增殖,且抑制率呈明显的剂量、时间依赖关系;两药联用作用24h时,表现为协同效应,而作用48h时主要表现为相加效应;槐耳清膏与DDP联用能显著增加MKN45细胞的凋亡率,并诱导细胞阻滞于S期。结论:槐耳清膏不仅能直接抑制胃癌细胞MKN45的增殖,与DDP联用时还能增加胃癌细胞对化疗的敏感性,而这种增敏作用可能是通过增加诱导细胞凋亡和S期阻滞而实现的。Objective： To observe the effect of Huaier Cream in enhancing chemosensitivity of human gastric carcinoma cells line MKN45 to DDP in vitro and to explore the possible mechanism. Methods： MTT was used to observe inhibitory effect of different concentrations Huaier cream and DDP on the growth of MKN45 ceils alone or combined, The combined effect and the interaction between Huaier cream and DDP were evaluated through calculating Q values. Flow cytometry was used to investigate the apoptosis and cell cycle distribution in the human gastric carcinoma cells line MKN45. Results： Huaier Cream or DDP could inhibit the growth of MKN45 cells in time and dose-dependent manner. The combination of the two drugs after 24h showed a synergistic effect, while it was mainly additive effect after 48h. The combined use of Huaier Cream and DDP could remarkably increase the apoptosis rate of MKN45 cells and induce cells accumulated in S phase. Conclusion： Huaier Cream could not only suppress the proliferation of MKN45 directly, but also enhance the sensitivity of MKN45 cells to DDP. The enhancement effect might be achieved by inducing apoptosis and S phase accumulation.
