摘 要:目的:研究经前期综合征(PMS)肝气逆证的发病机制及白香丹胶囊治疗该病证的作用靶点,为新药研发奠定基础。方法:采用电刺激应激法复制PMS肝气逆证大鼠模型,5d后进行脑组织固定及海马分离,用免疫荧光双标记技术检测GABABR1和GABABR2在各组大鼠海马中的分布,计算海马CA1和CA3区平均荧光强度(MFI),Western Blot检测ERK1/2、P-ERK1/2在各组大鼠海马中的表达,计算P-ERK1/2的相对含量。结果:免疫荧光双标记结果显示,各组大鼠海马中GABABR1和GABABR2分布模式无统计学差异;模型大鼠海马CA1和CA3区的GABABR1和GABABR2表达明显下降(P<0.01),海马P-ERK1/2的相对含量明显下降(P<0.01);给予白香丹后,GABABR1和GABABR2表达明显上升(P<0.05,P<0.01),海马P-ERK1/2的相对含量明显上升(P<0.01)。结论:海马CA1、CA3区GABABR亚基蛋白以及海马中ERK1/2蛋白水平下降可能是PMS肝气逆证的重要病机,也是白香丹胶囊的作用靶点。Objective: To study the pathogenesis of pre-menstrual syndrome (PMS) with liver qi invasion syndrome, and the targets of Baixiangdan capsule in treating PMS, to lay the foundation for the development of new medicine. Methods: The rat model of PMS with liver qi invasion syndrome was established by electric stimulation. Five days later, the brain tissue and separated hippocampal tissue were fixed, the distribution of GABABR1 and GABABR2 were detected by immunofluorescence double labeling technique in each group. The MFI was calculated in hippocampal CA1 and CA3 area. The expressions of ERK1/2, P-ERK1/2 in hippocampus of rats were detected in each group by Western Blot method, to calculate the relative content of P-ERKI/2. Results: The results of immunofluorescence double labeling showed that there was no significant difference in the distribution of GABABR1 and GABABR2 in hippocampus of rats in each group. The expressions of GABABR1 and GABABR2 in hippocampal CA1 and CA3 area were decreased obviously in the model group, the relative content of P-ERKI/2 was decreased obviously (P〈0.01). After the treatment of Baixiangdan Capsule, the expression of GABABR1 and GABABR2 were increased obviously (P〈0.05 or P〈0.01), the relative content of P-ERK1/2 was increased obviously (P〈0.01). Conclusion: The GABABR subunit protein and ERK1/2 protein levels decrease in hippocampal CA1 and CA3 area which may be an important pathogenesis of PMS with liver qi invasion, which are the targets of Baixiangdan Capsule.
