摘 要:目的:观察白术内酯Ⅰ、Ⅱ、Ⅲ对IEC-6细胞及食管癌细胞ECA9706增殖能力的影响。方法:细胞种植于96孔板,24h后分别加入白术内酯Ⅰ、Ⅱ、Ⅲ200、100、50、25、12.5、6.25μg/m L,放置培养箱48h后,MTT法检测白术内酯Ⅰ、Ⅱ、Ⅲ不同剂量对IEC-6细胞及ECA9706细胞增殖能力的影响。结果:白术内酯Ⅰ和白术内酯Ⅲ随着剂量的增加,呈现从增殖到抑制的作用,其中以白术内酯Ⅰ50μg/m L对IEC-6细胞增殖率效果最佳,白术内酯Ⅱ小剂量对IEC-6细胞作用不明显,但是随着剂量的增加,呈现抑制作用;白术内酯Ⅰ、Ⅱ、Ⅲ200、100、50μg/m L均能抑制ECA9706细胞的增殖,其中以白术内酯Ⅱ效果明显。结论:白术内酯Ⅰ可能是白术健脾的有效成分,而白术内酯Ⅱ可能是抑制肿瘤细胞增殖的主要成分之一。Objective: To observe the proliferation effect of atractylenolide Ⅰ, Ⅱ, Ⅲ on IEC-6 cells and ECA9706 cells. Methods: IEC-6 cells and ECA9706 cells were seeded in 96-well plates, and after 24 hours, respectively added atractylenolide Ⅰ, Ⅱ, Ⅲ at concentrations of 200, 100, 50, 25, 12.5, 6.251ag/mL, then continued to incubate for 48h, the proliferation ability of IEC-6 cells and ECA9706 cells were detected by MTT assay. Results: Atractylenolide I and HI showed different action from proliferation to suppress with increasing doses, atractylenolide at 50μg/mL had the best proliferation effect on IEC-6 cells, small dose of atractylenolide Ⅱ had no obvious effect on IEC-6 cells, it showed inhibition effect when the dose increased. Atractylenolide Ⅰ, Ⅱ, Ⅲ at 200, 100, 50μg/mL could inhibit the proliferation of ECA9706 cells. And the effect of atractylenolide Ⅱ was obvious. Conclusion: Atractylenolide I may be the main active ingredient in strengthening the spleen, atractylenolide Ⅱ may be one of the main ingredients in inhibiting tumor cells proliferation.
