摘 要:目的建立同时测定金线草中β-谷甾醇和胡萝卜苷的HPLC方法。方法采用HPLC-ELSD法,色谱柱以十八烷基硅烷键合硅胶为填充剂(250 mm×4.6 mm,5μm),甲醇—水(96∶4)为流动相洗脱,体积流量1.0 mL·min^-1,柱温30℃,漂移管温度70℃,载气体积流量1.8 L·min^-1,进样体积10μL。结果β-谷甾醇和胡萝卜苷分别在0.152 8-1.910μg(r=0.999 5)和0.101 6-1.270μg(r=0.999 3)有良好的线性关系;平均回收率为99.7%和98.9%,RSD分别为1.4%和1.8%(n=5)。结论该方法能准确测定金线草中β-谷甾醇和胡萝卜苷的含量。Objective To establish an ELSD-HPLC method for the simultaneous determination of β-sitosterol and daucosterol in the herb of Antenoron filiforme. Methods HPLC-ELSD method was adopted with C18column( 250 mm × 4. 6 mm,5 μm) and of methanolwater( 96∶ 4) as mobile phase. The flow rate was 1 mL·min^- 1,column temperature was 30℃,the temperature in drift tube was 70℃,the carrier gas flow was 1. 8 L·min^- 1and the introduction volume was 10 μL. Results The linear ranges of β-sitosterol and daucosterol were 0. 152 8 - 1. 910 μg( r = 0. 999 5) and 0. 101 6 - 1. 270 μg( r = 0. 999 3). The average recovery was 99. 7% and 98. 9%. RSD( n = 5) were 1. 4% and 1. 8%. Conclusions This method is accurate and reliable for the determination of the above two constituents in the herb of Antenoron filiforme.
