摘 要:目的观察丹红注射液对大鼠50%减体积原位肝移植术后存活及酶活性的影响,为临床合理用药提供理论依据。方法选择30对大鼠,随机分为模型组、丹红小剂量组、丹红大剂量组,采用改良双袖套法建立50%大鼠同种异体减体积原位肝脏移植动物模型,另设10只为空白对照组,其中丹红小剂量组、丹红大剂量组经尾静脉注射丹红注射液治疗14d,测定治疗后各组大鼠肝微粒体5种细胞色素P450亚型酶(CYPIA2、CYP2C9、CYP2D6、CYP2E1、CYP3A4)活性及治疗前后谷丙转氨酶(ALT)、谷草转氨酶(AST)和胆红素(TBil)水平,统计肝移植术受体大鼠存活率。结果丹红小剂量组、丹红大剂量组受体大鼠肝微粒体CYPIA2、CYP2C9、CYP3A4酶活性及ALT、AST水平均明显降低,受体大鼠存活率为70%,与模型组比较差异均有统计学意义(P均<0.05),丹红小剂量组、丹红大剂量组组间比较差异无统计学意义。结论丹红注射液可提高50%减体积原位肝移植大鼠的存活率,抑制大鼠肝微粒体CYPIA2、CYP2C9、CYP3A4酶活性,促进肝功能恢复。Objective It is to observe effect of Danhong injection onhepatic viability and enzyme activity in rats after 50%reduced size orthotopic liver transplantation,and provide a theoretical basis for clinical rational drug use.Methods 30 rats were selected and randomly divided into model group,small dose Danhong group,high dose Danhong group,using modified double cuff method to establish animal model of 50% rat orthotopic reduced size orthotopic liver transplantation,another 10 rats were chosen as blank control group.Small dose Danhong group andhigh dose Danhong group were treated with Danhong Injection by the tail vein injection for 14 days.Before and after treatment,the activities of 5 kinds of subtypes of enzyme in rat liver microsomal cytochrome(CYPIA2,CYP2C9,CYP2D6,CYP2E1,CYP3A4),the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST) and total bil TMGubin(TBil) were determined,survival rate of liver transplantation recipient rats were counted.Results The activities of rat liver microsomal CYPIA2,CYP2C9,CYP3A4 and the levels of ALT,AST were lower in small dose Danhong group andhigh dose Danhong grouop than those in model group,the survival rate was 70% in Danhong Injection groups,compared with the model group,the differences were all statistically significant(P all 〈0.05),but there was no significant difference between the two Danhong Injection groups.Conclusion Danhong Injection can increase survival rate of the rats after 50% reduced size orthotopic liver,inhibit the activities of CYPIA2、CYP2C9、CYP3A4 in rat liver microsomes,promote the recovery of liver function.
