摘 要:目的研究西红花苷提前用药对高海拔低氧条件下大鼠海马胰岛素样生长因子1(IGF-1)表达的影响。方法 224只SD大鼠,随机分为对照组和用药组。用药组每天肌肉注射西红花苷1次(50mg/Kg),对照组每天肌肉注射相同剂量的0.9%Nacl,连续给药3天后运至高海拔低氧环境,在第1、3、5、7天观察取材。通过Morris水迷宫观察大鼠学习记忆行为,通过免疫组织化学方法检测IGF-1的蛋白水平,通过RT-PCR方法检测IGF-1的mRNA水平。结果与对照组相比,用药组大鼠在Morris水迷宫实验中的寻台潜伏期在第1、3、5天明显缩短(P<0.05)。免疫组织化学法结果显示,用药组各组大鼠海马的IGF-1蛋白表达水平均高于对照组,其中以第1、3天最为明显(P<0.01)。RT-PCR法检测显示用药组各组大鼠海马的IGF-1 mRNA水平均高于对照组,其中以第1、3天最为明显(P<0.05)。结论西红花苷提前干预给药,可以提高高海拔低氧条件下大鼠海马IGF-1的表达量,同时能够改善高海拔低氧条件下大鼠学习记忆能力,其机制可能是通过调节IGF-1表达水平实现。Objective To investigate the effect of Crocin on the expression of insulin- like growth factor 1( IGF- 1) in hippocampus under hypoxia at high altitude in rats. Methods 224 SD rats were randomly divided into two groups: control and treatment group. The SD rats in treatment group were injected Crocin once daily( 50 mg / Kg)and the control group were injected the same dose of 0. 9% Nacl for 3d,and then they were shifted to high altitude under hypoxia environment and then the samples were taken at 1 d,3 d,5 d,7 d. By means of Morris water maze experiment to observe learning and memory behaviors of rats. The level of IGF- 1 in protein was detected by Immunohistochemical technology and the level of IGF- 1 in mRNA was detected by RT- PCR technology. Results Compared with the control group,treatment group showed significant less time in looking for platform in Morris water maze experiment at 1 d,3 d and 5 d( P〈 0. 05). The results of immunohistochemical showed that the protein expression level of IGF- 1 in hippocampus of rats was higher in treatment group than the control group,and the expression levels in 1d and 3d were obviously higher( P〈 0. 01). RT- PCR showed that the mRNA level of IGF- 1in hippocampus of rats was higher in treatment group than that of in control group,and the expression level in 3d were obviously higher( P〈 0. 05). Conclusions Crocin pretreatment can increase the expression level of IGF- 1 in hippocampus of rats under the hypoxic condition at high altitude. Crocin shows the effect of protecting brain function and improving learning and memory ability of rats under hypoxia at high altitude,and its mechanism may be related with the effect of modulating the IGF- 1 expression.
