摘 要:目的观察高海拔环境下大鼠脑组织海马区的细胞形态结构变化和S100B蛋白的表达,探索缺氧刺激后脑组织损伤和修复的机制。方法 120只健康成年SD大鼠,随机分成两组:模拟海拔5 000米组(实验组:H组)和海拔2 200米组(对照组:M组);H组经模拟海拔5 000米低压氧舱预处理3天,转移至海拔2 200米环境下饲养,M组不经模拟海拔5 000米低压氧舱预处理,直接在海拔2 200米环境饲养。两组又分为缺氧损伤后较低海拔环境恢复1、3、7、14、28天五个亚组,在相应时间点取脑组织,分别用Nissl、免疫组织化学染色方法染色,显微镜下观察海马区细胞结构,进行阳性细胞计数,并对相关结果进行分析。结果 Nissl染色可见M组大鼠海马区神经元排列较整齐、结构完整,未见明显损伤细胞;H组大鼠海马区神经元细胞数量减少、排列紊乱,其中H组1天和3天亚组出现明显的组织水肿,出现变性和核浓缩的损伤细胞;S100B免疫组化染色阳性细胞计数结果显示,H组S100B蛋白的表达在低压缺氧刺激后第1、3天明显高于M组对应时间点(P<0.05),其他时间点S100B蛋白表达量无统计学差异。结论高海拔低压缺氧环境下高表达的S100B蛋白诱导大鼠海马细胞形态和组织结构损伤。Objective To observe the hippocampus structure change and S100 B protein expression under high altitude environment,to explore the mechanisms of brain injury and restore after hypobaric hypoxia. Methods 120 SD rats were randomly divided into 2 groups: group H: high altitude group( 5000m); group M: control group( 2200m). Group H was pretreated 3 days in the low pressure oxygen tank to simulate the 5 000 meters environment,then shift them to 2 200 meters environment. Both group were divided into five subgroup after hypoxia injury:1d,3d,7d,14 d,and 28 d,respectively. Nissl staining and immunohistochemical staining to detect the hippocampus structure and positive cell count. Results Nissl staining in group M showed that the hippocampal neurons arranged neatly and structural integrity,no obvious damage cells; Group H was observed that the number of neurons decreased and disordered arrangement in hippocampus and subgroup 1d and 3d appeared obvious tissue edema; for the S100 B protein positive cell counting,after low pressure of hypoxia stimulation 1d or 3d,the positive cell number was significantly higher than that of group M with corresponding time points( P〈 0. 05). Conclusion S100 B protein is expressed higher level under hypobaric hypoxian and the increased S100 B protein may induce the morphological change and cell damage in hippocampus of rats.
