摘  要:目的:研究滋肾清肝降糖方(ZQJC)对2型糖尿病(T2DM)大鼠糖代谢、蛋白酪氨酸磷酸1B(PTP1B)表达及超氧化物歧化酶(SOD)、丙二醛(MDA)和游离脂肪酸(FFA)的影响。方法:70只雄性SD大鼠随机抽取10只大鼠为正常组,其余大鼠采用高脂饲料喂养联合ip 30 mg·kg-1链脲佐菌素(STZ)的方法造T2DM模型,分为模型组、二甲双胍组(Met,0.14 g·kg-1·d-1,ig)、滋肾清肝降糖方低剂量组(0.9 g·kg-1·d-1,ig)、滋肾清肝降糖方中剂量组(1.8 g·kg-1·d-1,ig)及滋肾清肝降糖方高剂量组(3.6 g·kg-1·d-1,ig)。检测药物干预2,4周后各组大鼠空腹血糖(FBG)以及4周后各组大鼠血清中SOD,MDA,FFA的水平及肝细胞匀浆中PTP1B蛋白表达。结果:与正常组比较,模型组的FBG均升高(P<0.01),PTP1B在肝中的表达增加(P<0.01);与模型组比较,ZQJC各剂量组可显著降低大鼠血清FBG,FFA和MDA的含量(P<0.01,P<0.05),升高血清SOD的活性(P<0.05),ZQJC可降低PTP1B在肝中的表达。结论:滋肾清肝降糖方对2型糖尿病大鼠有调节糖代谢的作用,抑制PTP1B表达及氧化应激反应可能为其作用机制之一。Objective: To study the effect of Zishen Qinggan Jiangtang compound (ZQJC) on glucose, protein tyrosine phosphatase 1B (PTP1B) expression, superexide dismutase (SOD), malondialdehyde (MDA) and free fatty acide (FFA) in type 2 diabetic mellitus (T2DM) rats. Method: Ten rats were randomly selected as normal group, type 2 diabetes model was induced with high sucrose-fat diet and STZ ip for 30 mg·kg^-1, rats were randomly divided into groups: model group, metformin (Met) group (0. 14 g ·kg^-1·d^-1, ig), ZQJCL group (0.9 g·kg^-1·d^-1, ig), ZQJCM group (1.8 g·kg^-1·d^-1, ig) and ZQJCH group (3.6 g·kg^-1·d^-1, ig), 10 rats in each group, fasting blood glucose (FBG) was detected at 2^rd week and 4^th week, and the level of SOD, MDA, FFA in the rats' serum and PTP1B expression in liver at 4^th week were measured after the treatment. Result: Compared with the normal group, the blood glucose levels of model group were increased (P 〈 0.01 ), PTP1B expressions in liver of model group were increased (P 〈 0.01 ). After treatment, compared with model group, ZQJC group could obviously decrease the levels of FBG, MDA and FFA (P 〈 0.05, P 〈 0.01 ), improve SOD activity (P 〈 0.05) and inhibit the PTP1B expression in liver. Conclusion: ZQJC has the effect of regulating glucose metabolism of the T2DM rats, inhibition of oxidative stress and PTP1B expression in liver could be one of the mechanisms.
