摘  要:以玉米(Zea mays L.)为材料,通过酶联免疫法、实时荧光定量PCR技术、免疫共沉淀激酶反应以及原生质体瞬时表达体系等方法研究了在油菜素内酯(Brassinosteroids,BR)诱导的玉米叶片抗氧化防护过程中钙调素(Calmodulin,Ca M)的作用以及Ca M和Ca2+/Ca M依赖的蛋白激酶(Zm CCa MK)之间的关系。结果显示:外源BR处理显著提高了玉米叶片叶肉细胞原生质体中的Ca M含量,Ca M拮抗剂的预处理几乎完全抑制了BR诱导的抗氧化防护酶活性,表明Ca M参与了BR诱导的抗氧化防护。而且Zm CCa MK原生质体瞬时表达及瞬时沉默结果显示Zm CCa MK影响BR诱导的Ca M含量,Ca M拮抗剂的预处理也显著抑制了BR诱导的H2O2的产生、Zm CCa MK基因表达以及激酶活性。说明BR诱导产生的Ca M增强H2O2的积累,进一步诱导抗氧化防护酶活性增加,并且Ca M和Zm CCa MK之间存在互动机制。In this study,enzyme-linked immunosorbent assay( ELISA),real-time PCR,immunoprecipitation and in-gel kinase assay and protoplast transient expression system were used to investigate the role of calmodulin( Ca M) in brassinosteroids( BR)-induced antioxidant defense and the relationship between Ca M and Ca2 +/ Ca M-dependent protein kinase( Zm CCa MK) in leaves of maize( Zea mays L.) plants. The results showed that treatment with BR led to significant increase in the content of Ca M in protoplasts of maize plants and pre-treatment with Ca M antagonists blocked the activities of antioxidant enzymes induced by BR treatment in leaves of maize,which suggest that Ca M is involved in BR-induced antioxidant defense. Moreover,transient expression or transient silencing of Zm CCa MK in protoplasts resulted in a significant enhancement or block in the content of Ca M induced by BR treatment. Pre-treatment with Ca M antagonists significant inhibited the production of H2O2,the expression and the activity of Zm CCa MK induced by BR in the leaves of maize. The results suggest that the BR-induced Ca M increased H2O2 accumulation,increased the activities of antioxidant enzymes, and a cross-talk may exist between Ca M and Zm CCa MK in BR signaling in the leaves of maize plants.
