摘 要:[目的]证实从蜡梅花基因组提取DNA等同于从叶基因组提取DNA。[方法]采用CTAB法,对保康野生蜡梅花与叶基因组进行遗传多样性研究。[结果]在用CTAB法提取DNA时,65℃水浴振荡并二次抽提后花基因组DNA在浓度和OD260/OD280上都与叶基因组DNA达到相近的水平。[结论]从性状表现更显著的蜡梅花基因组提取DNA进行遗传性分析能达到叶基因组的水平。[Objective] The aim was to confirm that Chimonanthus praecox genomic DNA extracted from the flower was equivalent to extract genomic DNA from the leaf. [Method] Using CTAB methods,genetic diversity of wild Chimonanthus praecox flower and leaf genome in Baokang County were studied. [Result] When DNA was extracted using CTAB methods,after vibration in 65 ℃ water bath and two extractions,concentration and OD260/ OD280 value of flower total genome DNA were at similar standard to those of leaf total genome DNA. [Conclusion]DNA extracted from the more significant traits Chimonanthus praecox flower genome could reach the level of the leaf genome.
