摘  要:目的研究突触囊泡融合蛋白Munc18-1在小鼠耳蜗内毛细胞的表达及定位。方法应用激光共聚焦成像技术,通过免疫荧光单标法检测Munc18-1蛋白在小鼠耳蜗内毛细胞中的表达,免疫荧光双标法检测其与带状突触蛋白Ct BP2的定位关系。结果通过激光共聚焦成像技术检测发现,免疫荧光单标染色显示内毛细胞胞质中有高浓度的Munc18-1蛋白表达,免疫荧光双标染色结果显示Munc18-1蛋白聚集在Ct BP2蛋白附近。结论 Munc18-1蛋白在耳蜗内毛细胞中有表达,聚集在突触蛋白Ct BP2周围,可能与维持突触囊泡融合、胞吐功能紧密相关。Objective To investigate the expression of Munc18-1 protein in the inner hair cells of mouse cochlea and identify its location. Methods Thirty C57 mice( 60 cochleae) were randomly detected with Munc18-1 antibody or Ct BP2 + Munc18-1 double antibodies by immunofluorescence assay and confocal laser imaging technique to detect the expression and location of Munc18-1 protein in the cochlea inner hair cells. Results Laser scanning confocal imaging indicated that the Munc18-1 protein was highly expressed in the cytoplasm of the inner hair cells,and Munc18-1 protein was aggregated close to the synaptic protein Ct BP2. Conclusion Munc18-1 protein exists in the mouse cochlea inner hair cells,and aggregates with synaptic protein Ct BP2. It might be closely involved in the maintenance of synaptic vesicles fusion and exocytosis.
