摘 要:目的观察重复阳极经颅直流电刺激(transcranial direct current stimulation,t DCS)对阿尔茨海默病(Alzheimer’s disease,AD)大鼠学习记忆能力的影响。方法将36只SD大鼠分为假手术组、AD组、20μA刺激组、60μA刺激组、100μA刺激组和200μA刺激组,每组6只。AD组和刺激组的海马注射Aβ1-40制作AD模型。术后2周,刺激组接受重复阳极t DCS,而AD组和假手术组接受假刺激。t DCS后,用Morris水迷宫和Y迷宫检测大鼠学习记忆能力,尼氏染色观察大鼠海马神经元形态变化。结果 Morris水迷宫结果显示,60、100、200μA刺激组的潜伏期均较AD组明显降低(P<0.05)。大鼠经过原平台的次数和原平台象限游泳时间各组间差异并没有统计学意义(P>0.05)。Y迷宫结果显示100μA和200μA刺激后,大鼠学习记忆成绩较AD组均有明显改善(P<0.05)。尼氏染色显示t DCS后大鼠海马神经元胞质中尼氏体数量增加,染色加深。结论重复阳极t DCS能改善AD大鼠学习记忆功能障碍。Objective To investigate the effect of repeated anodal transcranial direct current stimulation( t DCS) on the learning and memories abilities in rat model of Alzheimer 's disease( AD).Methods Sprague-Dawley rats were randomly allocated to a sham group,an AD group,a 20 μA t DCS group,a 60 μA t DCS group,a 100 μA t DCS group and a 200 μA t DCS group. The hippocampi of the rats in the AD group and the t DCS groups was subjected to Aβ1- 40 injection to establish AD models. In postoperative2 weeks,repeated anodal t DCS was applied to the frontal cortex of the rats in the t DCS groups,while the rats in the AD group and the sham group received sham stimulation. After t DCS,the learning and memory abilities of the rats was assessed by Morris water maze and Y-maze tests,and Nissl staining was performed to reveal morphological changes of hippocampus neurons. Results The Morris water maze test results showed that,compared with the AD group,the 60 μA t DCS group,100 μA t DCS group and 200 μA t DCS group showed significant decrease of the escape latency( P〉0. 05),but there were no significant changes in the platformcrossing times and swimming time in platform quadrant( P〈0. 05). The Y-maze test results showed that,t DCS with current intensity of 100 μA and 200 μA significantly improved the learning and memory performance of the AD rats( P〈0. 05). t DCS could increase the Nissl body number and deepen staining.Conclusion Repeated anodal t DCS may alleviate learning and memory dysfunction in the rat models of AD.
