异丙酚对发育期小鼠大脑皮质锥体细胞成熟和树突发育的影响

摘  要:目的探讨异丙酚对小鼠大脑皮质发育阶段神经元成熟和树突发育的影响。方法选用同窝的发育期小鼠按随机数字表法分为3组:1%脂肪乳剂注射对照组,30 mg异丙酚注射组,60 mg异丙酚注射组。于出生后第7天(P7)经腹腔注射。采用免疫荧光方法检测异丙酚注射注射对P7小鼠大脑皮质内胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)免疫反应阳性的星形胶质细胞及Neu N免疫反应阳性的成熟神经元的变化,MAP2B免疫组化检测锥体细胞形态与数量的变化。运用高尔基染色观察异丙酚对P14小鼠大脑皮质内锥体细胞树突棘总数及亚型数量的变化。结果不同剂量异丙酚对P7小鼠大脑皮质星形胶质细胞和成熟神经元细胞数量均无显著影响,MAP2B免疫组化显示60 mg异丙酚注射显著减低P7小鼠大脑皮质内锥体细胞的数量与树突总数,而30 mg异丙酚注射则无显著改变。高尔基染色结果进一步证实:与对照组比较,60 mg异丙酚注射显著减少P14小鼠大脑皮质锥体细胞树突棘总数(P<0.05)及蘑菇样树突棘数量(P<0.05);30 mg异丙酚注射亦显著减少蘑菇样树突棘数量(P<0.05),而对树突棘总数的影响未达到统计学差异。结论异丙酚单次注射对发育期小鼠大脑皮质成熟神经元及星形胶质细胞数量影响不明显,但高剂量注射对皮质锥体细胞树突的发育与树突棘的成熟有显著抑制作用。Objective To determine the effect of propofol on the maturation and dendritogenesis of neurons in the developing cerebral cortex. Methods Mice from the same litters were randomly divided into3 groups with 6 rats in each group,that is,control group( 1% intralipid),low-dosage propofol group( 30 mg /kg propofol) and high-dosage propofol group( 60 mg /kg propofol). The corresponding drugs were injected intra-peritoneally in 7 d after born( P7). In 1 h after the injection,immunofluorescence assay was used to detect the expression of glial fibrillary acidic protein( GFAP) and neuronal nuclei( NEUN) in the cerebral cortex of mice,and microtubule-associated protein 2 B( MAP2B) was used to detect the changes in the morphology and the number of cortical pyramidal cells. In 7 d after the injection( P14),Golgi staining was further used to analyze the numbers of the total dendritic spines and its subtypes of cortical pyramidal cells. Results No significant difference was observed in GFAP-positive cells and NEUN-labeled cells in the cerebral cortex between mice treated with different dosage of propofol at P7. The MAP2 B immunochemistry confirmed that the number of cortical pyramidal cells and its total dendrites were significantly lower in the highdosage group than in control group at P7,while no significant difference was observed between the low-dosage group and control. Moreover,Golgi staining further demonstrated that the number of total dendritic spines of cortical pyramidal cells( P〈0. 05) and the number of mushroom-type spines( P〈0. 05) were significantly lower in the high-dosage group at P14 than the control. No significant difference was observed in the number of total dendritic spines while the number of mushroom-type spines was significant reduced( P〈0. 05) in lowdosage group compared with control group.
