摘  要:目的研究补肾益精代表方左归丸对衰老大鼠骨髓间充质干细胞(MSC)形态学方面的影响。方法通过腹腔注射D-半乳糖造模,制备衰老大鼠模型,分离培养骨髓MSC。光镜观察各组大鼠MSC一般形态学特征,电镜观察其超微结构特点。结果光镜观察模型组P3代大鼠骨髓MSCs呈现衰老细胞形态学特征,左归丸组大鼠BMSCs的细胞形态与正常组相近。透射电镜观察显示,模型组大鼠P3代骨髓MSCs细胞内结构改变呈现衰老特征,左归丸中剂量组大鼠的骨髓MSCs电镜下的超微结构与正常组最相近。结论衰老大鼠骨髓MSCs的细胞形态各方面表现有明显改变,左归丸水提液在一定浓度范围内能延缓大鼠骨髓MSCs的衰老。Objective To investigate the effect of Zuogui pill on aging rats bone marrow mesenchymal stem cell( MSC) morphology. Methods Aging rat model was made by intraperitoneal injection of D- galactose. MSCs were stained by Giemsa method,and were observed by the optical microscope and electron microscope respectively. Results Under the optical microscope,MSC of model group presented morphological characteristics of senescent cells,while those in Zuogui pill groups were close to the normal group. Under the electron microscope,MSC of model group presented senescent cells morphological characteristics,but the ultrastructure of the Zuogui pill groups BMSCs were close to the normal group. Conclusion The morphology of aging rat BMSC have obvious change,Zuogui pill which in a certain concentration range can delay the aging of rat BMSC.
