中药枳壳挥发油成分气相色谱-质谱联用分析和促进胃肠动力药效研究

摘　　要：目的明确枳壳挥发油的主要成分及各成分的相对含量,研究其促胃肠动力活性。方法采用药典方法提取枳壳挥发油,用气相色谱-质谱法对其化学成分进行分析,经计算机处理和NIST298、WIL EY275标准质谱图库检索鉴定各种化学成分,并采用归一法测定各组分的百分含量。采用半固体糊碳末推进法,以胃残留率和小肠推进率为指标,考察枳壳挥发油促胃肠动力作用。结果共鉴定出15个挥发油成分,其中主要成分为柠檬烯,相对含量为64.524%。且枳壳挥发油和柠檬烯均有显著的促胃肠动力作用。结论枳壳挥发油主要为萜类成分,是其发挥促胃肠动力作用的主要药效成分之一,其中柠檬烯的含量与枳壳的促胃肠动力作用呈一定的正相关性。该研究为揭示枳壳药材促胃肠动力物质基础和作用机制提供了可靠的科学依据。Objective To figure out the main components of the orange essential oil and the relative content of each component,and to research the prokinetic effect of them on the stomach and intestine. Methods Pharmacopoeia method is used to extract orange essential oil,analyse the components by GC- MS,processed by computer and match with NIST298,WIL EY275 standard mass spectrometry gallery,then measure the percentage of each component by area normalization method. Gastric residual rate and small intestinal propulsion rate were taken as the comprehensive indexes to research the prokinetic effect of the orange essential oil by the method of semisolid paste carbon propulsion. Results 15 kinds of volatile oil components were identified,the main component is limonene,and the relative content of it is 64. 524 %. The orange essential oil and the limonene have significant prokinetic effect on the stomach and intestine. Conclusion The terpene are the main ingredients of the orange essential oil and one of the medicinal compositions on the prokinetic effect,the content of limonene and the prokinetic effect of fructus aurantii are positively related to the straight line. This study will provide material base on the prokinetic effect of fructus aurantii and a reliable scientific basis on its mechanism.
