人参皂苷Rb1与小檗碱配伍对糖尿病小鼠糖脂代谢的影响  被引量:16

摘  要:目的观察人参皂苷Rb1(Rb1)与小檗碱(BBR)配伍对肥胖糖尿病小鼠糖脂代谢的影响。方法将db/db小鼠分为4组,分别为模型组(db/db组),人参皂苷Rb1组(Rb1组),小檗碱组(BBR组),人参皂苷Rb1和小檗碱配伍组(Rb1+BBR组),另设正常对照组(正常组)。Rb1组和BBR组分别按照20mg·kg-1体重腹腔注射Rb1和BBR,Rb1+BBR组按照20mg·kg-1体重同时腹腔注射Rb1和BBR,给药14 d后,取血测定血糖、胰岛素、血脂以及肿瘤坏死因子-α(TNF-α)和脂联素水平,此外,检测肝脏甘油三酯含量。结果与db/db组比较,Rb1和小檗碱明显降低肥胖糖尿病小鼠的胰岛素水平和胰岛素抵抗指数(HOMA-IR)(P<0.05),同时降低血清胆固醇含量和游离脂肪酸含量(P<0.05)以及肝脏内TG含量,Rb1和小檗碱联合治疗后,相比Rb1组或者BBR组,以上糖脂代谢相关指标未见进一步下降;BBR组糖尿病小鼠的摄食量和体重以及附睾脂肪和肝脏重量明显减少(P<0.05或P<0.01),而Rb1组仅见肝脏重量显著下降(P<0.05),与BBR组比较,Rb1+BBR组的摄食量和体重以及附睾脂肪和肝脏重量未见显著性差异;Rb1组、BBR组和Rb1+BBR组,与db/db组比较,血清脂联素水平升高(P<0.05),血清TNF-α水平降低也有不同程度的下降,但是三组之间无显著性差异。结论人参皂苷Rb1和小檗碱均能明显改善糖尿病小鼠的胰岛素抵抗和糖脂代谢紊乱,但是两者配伍使用未见到显著的增效作用。Objective To observe the effects of combination of ginsenoside Rb1( Rb1) and berberine( BBR) on glucose and lipid metabolism in db / db obese diabetic mice. Methods Male db / db mice were randomly divided into four groups: diabetic control group( db / db group),Rb1 group,BBR group,and Rb1 + BBR group,male C57 BLKS / J mice were observed as normal control group( normal group). Mice in Rb1 group and BBR group were administered with Rb1 or berberine by intraperitoneal injection at the dosage of 20mg·kg- 1,respectively. Mice in Rb1 + BBR group were intraperitoneally injected with combination of Rb1 and BBR at the dosage of 20mg·kg- 1. After 14 days treatment,serum was collected for measurements of fasting glucose( FBG),insulin( FINS),blood lipid,free fatty acid( FFA),TNF- αand adiponectin. In addition,liver triglyceride content was examined.Results Comparing to that in db / db group,FINS,HOMA- IR,total cholesterol,FFA and liver TG were lower significantly in Rb1 group and BBR group. However,these parameters did not show further decrease in Rb1 + BBR group. Food uptake,body weight,epididymal adipose tissue and liver weight were reduced in BBR group,but only liver weight reduced in Rb1 group. There is no significant difference in food uptake,body weight,epididymal adipose tissue and liver weight between BBR group and Rb1+ BBR group. TNF- α was reduced and adiponectin was elevated after treatment with Rb1,BBR or Rb1 + BBR,but which were not obvious different in three group. Conclusion Ginsenoside Rb1 and berberine could improve abnormal metabolism of glucose and lipid in diabetic mice,combination of ginsenoside Rb1 and berberine does not produce synergistic effect.
