摘  要:目的观察大黄虫丸对慢性粒细胞性白血病K562细胞抑制增殖、诱导凋亡及对PI3K/AKT表达的影响,进而探讨其治疗慢性粒细胞性白血病的作用机制。方法制备大黄虫丸含药血清,应用MTT法检测对K562细胞增殖的影响,细胞克隆形成实验检测对细胞克隆形成的影响,用Annexin V FITC/PI法检测对细胞凋亡和细胞周期的影响,免疫印迹法检测PI3K/AKT信号通路重要蛋白Akt及其磷酸化形式p-Akt的蛋白水平。相关实验数据用SPSS17.0统计软件进行统计学处理。结果大黄虫丸含药血清能显著抑制白血病K562细胞的增殖,且呈浓度、时间依赖性;能抑制K562细胞克隆的形成;能诱导K562细胞的凋亡,且将细胞阻滞于G0/G1期,呈浓度依赖性。随着浓度升高,p-Akt/Akt值呈降低趋势,差异有统计学意义(P<0.05)。结论大黄虫丸能够体外抑制K562细胞增殖和诱导其凋亡,可能通过抑制PI3K/AKT信号通路激活实现。Objective explore the influences of rhubarb zhechong pill on proliferation,apoptosis and PI3 K / Akt signaling pathway in K562 cells. Methods Effects of the rhubarb Zhechong pill on the K562 cell proliferatio and apoptosis were determined by MTT assay and cell colony formation assay,K562 cell cycle and apoptotic percentage were detected by flow cytometry. Results The rhubarb Zhechong pill could significantly inhibit the proliferation of K562 cells in a time and concentration dependent manner; It can inhibit the formation of K562 cell clones; In addition,it can induce the apoptosis of K562 cells,and the cells were blocked in G0 /G1 phase in a concentration- dependent manner. The p- Akt / Akt value decreased with increasing concentration and the differences were statistically significant among concentrations( P 0. 05). Conclusion The rhubarb Zhechong pill can inhibit the proliferation and apoptosis of K562 cell possibly by inhibiting the activation of PI3 K / Akt pathway activation.
