摘 要:目的观察脑通汤对血管性痴呆大鼠学习记忆能力以及海马CA1区脑微血管密度(MVD)及血管场面积的影响。方法采用改良的4-VO法制备大鼠模型,脑通汤灌胃治疗后Morris水迷宫检测大鼠学习记忆能力,免疫组化法检测海马区CA1区微血管密度(MVD)及血管场面积。结果与模型组比较,脑通汤大、中剂量组大鼠逃避潜伏期明显缩短,穿越平台次数增加,微血管密度(MVD)及血管场面积明显增高(P<0.01)。与多奈哌齐组比较,脑通汤大、中剂量组MVD及血管场面积均显著增高,有统计学差异(P<0.01)。结论脑通汤可促进微血管新生,增加缺血区血液灌流,保护海马区NVU整体功能,从而改善VD大鼠学习记忆能力。Objective To investigate the ability of learning and memory,the microvessel density( MVD) and sum area of lumens changes in the hippocampus CA1 area of vascular dementia rats. Methods By using modified Pulsinelli 4 vascular occlusions method( 4- VO) preparation of vascular dementia( VD) model. Morris water maze determination of learning and memory ability in rats after 4 weeks of treatment with Naotong Decoction. Immunohistochemical method to detect the microvessel density( MVD)and sum area of lumens in the hippocampus CA1 area. Results Compared with the model group,the escape time were reduced and the times of spanning the previous platform were increased,and the microvessel density( MVD) and sum area of lumens were increased in Naotong Decoction large- dose and middle- dose treatment group( P 0. 01). Compare to Donepezil HCL group,The high dose and moderate dose groups can obviously heighten the microvessel density( MVD) and sum area of lumens( P 0. 01). Conclusion Naotong decoction can obviously heighten the microvessel density( MVD) and sum area of lumens,increase newly born microvessel and promote the establishment of collateral circulation in hippocampal area,and protect hippocampus neurovascular unit function,consequently improve learning and memory abilities of VD rats.
