摘  要:目的比较卫矛提取物不同极性组分总黄酮含量及抗氧化抗炎活性。方法 60%乙醇提取卫矛黄酮类化合物,经萃取制得4个不同极性的组分(石油醚组分、乙酸乙酯组分、正丁醇组分、水组分)。采用分光光度法测定各组分的总黄酮含量;通过清除DPPH自由基、ABTS自由基试验和还原力试验,评价各组分的抗氧化活性;利用脂多糖诱导小鼠腹腔巨噬细胞RAW264.7释放一氧化氮模型,评价各组分的抗炎活性。结果卫矛提取物总黄酮的含量为(271.25±5.51)mg·g-1,其中乙酸乙酯组分的总黄酮含量最高,为(773.61±12.03)mg·g-1,显著高于总提取物和其他组分(P<0.01)。活性评价结果表明卫矛提取物具有良好的抗氧化和抗炎活性,其中乙酸乙酯组分的抗氧化活性最强,乙酸乙酯组分和石油醚组分则具有良好的抗炎活性。结论卫矛的化学成分具有良好的多样性,其作用机制具有多组分、多靶点、多途径的特点,其具体机制有待进一步深入研究。Objective To compare the total flavonoids contents,antioxidant and anti- inflammatory activity of different fractions of Euonymus alatus( Thunb.) Sieb extraction. Methods The flavones were segregated from Euonymus alatus( Thunb.) Sieb with60% ethanol,and then extracted with petroleum ether,ethyl acetate,butanol,respectively. Contents of total flavonoids in various fractions were determined by spectrophotometric. Antioxidant activity of the different fractions were evaluated by DPPH,ABTS and reducing power measurement,and anti- inflammatory activity were evaluated by measuring NO that released from mice's abdominal macrophage RAW264. 7 induced by lipopolysaccharide. Results The content of total flavonoids of ethanol extraction of Euonymus alatus( Thunb.) Sieb. was 271. 25 ± 5. 51 mg·g- 1. The content of total flavonoids of ethyl acetate fraction was 773.61 ± 12. 03 mg·g- 1,which was significantly higher than any other fractions( P 0. 01). The results revealed that Euonymus alatus extraction has strong antioxidant and anti- inflammatory activity. All fractions had antioxidative and anti- inflammatory activity. Among them,the ethyl acetate fraction has the most potent antioxidative effect and the acetate fraction and petroleum ether fraction had the most potent anti- inflammatory effect. Conclusion The chemical components of Euonymus alatus showed good diversity. The molecular mechanisms of Euonymus alatus was featured by multi- drugs,multi- targets and multi- pathways. It is required further in- depth study.
