摘 要:目的:观察电针对APP/PS 1转基因小鼠β淀粉样蛋白(Aβ)及其清除途径的关键转运受体低密度脂蛋白受体相关蛋白-1(LRP 1)的可能影响,探讨电针干预阿尔茨海默病的可能途径。方法 :将4月龄APP/PS 1双转基因小鼠随机分为模型组、电针治疗组,以背景小鼠C 57BL/6为正常对照组。电针治疗组予电针"涌泉""百会",15min/次,隔日1次,治疗6周。采用Morris水迷宫测试小鼠空间记忆行为学,ELISA法检测海马Aβ1-40、Aβ1-42含量,Western blot法检测海马LRP 1表达。结果:Morris水迷宫测试显示,各组定位航行实验逃避潜伏时以及空间探索实验穿越平台象限次数、平台象限停留时长的差异没有统计学意义(P>0.05),模型组逃避潜伏时相比正常对照组有延长趋势,而电针治疗组逃避潜伏时较模型组有缩短趋势。模型组海马Aβ1-42、Aβ1-40含量及Aβ1-42/Aβ1-40值均显著高于正常对照组(P<0.01),电针治疗组Aβ1-42、Aβ1-40含量及Aβ1-42/Aβ1-40值均显著低于模型组(P<0.01)。模型组海马LRP 1蛋白相对表达量低于正常对照组(P<0.05),电针治疗组LRP 1相对表达量与模型组比较差异无统计学意义(P>0.05)。结论:电针可降低APP/PS 1双转基因小鼠海马Aβ水平,其机制是否与Aβ转运受体LRP 1相关尚需进一步探讨。Objective To observe the effect of electroacupuncture (EA) treatment on the level of hippocampal amyloid- beta peptide (Aβ) and its key transport receptor low density lipoprotein receptor-related protein-1 (LRP 1) in APP/PS I trans- genic mice so as to explore its mechanism underlying improvement of AIzheimer's disease (AD). Methods Twenty-four male APP/PS 1 transgenic mice were equally and randomly divided into model group and EA treatment group, and 12 C 57 BL/6 mice were used as the normal control group. EA (1 Hz/50 Hz, 0.3 mA) was applied to "Baihui" (GV 20) and "Yongquan" (KI 1) for 15 min, once every other day for 6 weeks. The learning-memory ability was detected by using Morris water maze testing, left hip- pocampal Aβ 1-40 and Aβ 1-42 contents were assayed by ELISA, and right hippocampal LRP 1 expression was detected using Western blot(WB). Results Results of Morris water maze test showed no significant differences among the three groups in the escape latency, the times of the platform-site crossovers, the time spent in the target platform quadrant (P~〉0.05). Compared with the model group, the moderately increased escape latency had a decreasing tendency in the EA treatment group, ELISA as- saying showed that hippocampal Aβ 1-42 , Aβ 1-40, and ratio of Aβ 1-42/Aβ 1-40 of the model group were significantly higher thanthose of the normal control group (P〈0.01). After EA intervention, the increased Aβ 1-42 , Aβ 1-40, and ratio of Aβ1-42/Aβ 1- 40 were remarkably down-regulated in the EA treatment group( P〈0.01 ). WB detection displayed that the right hippocampal LRP 1 expression level of the model group was markedly lower than that of the normal control group (P〈0.05). After EA treat- ment, LRP 1 expression level was moderately up-regulated but without significant difference between the model and EA treatment groups (P〉0.05).
