摘  要:目的研究新疆哈萨克族高血压患者外周血T淋巴细胞是否表达NLRP3炎症小体信号通路相关分子及其水平变化。方法随机选取2013年6月至2014年2月期间新疆医科大学第一附属医院心脏中心门诊首次就诊且未经降压药物治疗的新疆哈萨克族高血压患者30例(高血压组),年龄(54.7±5.6)岁,选取同期同民族健康体检者30例(对照组),年龄(54.6±4.7)岁,每组男女比例1∶1。采集两组受试者外周血并分别提取血浆和T淋巴细胞,采用酶联免疫吸附试验(ELISA)法检测血浆白细胞介素(IL)-1β含量;运用荧光定量逆转录聚合酶链反应(RT-PCR)和Western blot法检测T淋巴细胞中NLRP3、半胱氨酸天冬氨酸蛋白水解酶1(Caspase-1)、IL-1βmRNA和蛋白的表达。结果两组受试者基线资料差异无统计学意义(P>0.05)。与对照组相比,高血压组血浆IL-1β水平明显升高[(61.0±4.1)比(43.0±3.1)ng/L,P=0.002]。外周血T淋巴细胞表达NLRP3炎症信号通路关键分子NLRP3、Caspase-1和IL-1β;且高血压组的这3种基因mRNA相对表达量均高于对照组(0.0018±0.0005比0.0005±0.0001,P=0.010;0.0205±0.0062比0.0065±0.0021,P=0.024;0.0092±0.0019比0.0033±0.0011,P=0.008);三者蛋白表达量也较对照组增高。结论外周血T淋巴细胞上表达NLRP3炎症小体信号通路相关分子,并且该通路的相关分子在新疆哈萨克族高血压患者外周血T淋巴细胞上表达增多。Objective To explore whether NLRP3 inflammasome and its downstream molecules are expressed in pa- tients with essential hypertension (EH) in Xinjiang Kazakh and their expression changes. Methods Thirty un- treated Kazakh hypertensive patients [mean age (54.7±5.6) years] and 30 Kazakh healthy subjects [mean age (54.6±4.7) years] were randomly collected from June 2013 to February 2014 in Heart Center of the First Affiliated Hospital in Xinjiang. The male to female ratio was 1 : 1 in each group. The serum and peripheral blood T lym- phocytes were separated. Enzyme linked immunosorbent assay (ELISA) was performed to measure the serum level of interleukin (IL) -1β. The real time quantitative RT-PCR and Western blot analysis were used to detect the mRNA and protein expression levels of NLRP3 inflammasome and its downstream signaling pathway-related mole- cules, respectively. Results No significant differences in baseline data of the two groups was observed (P〉0.05). Results of ELISA revealed an elevated serum level of IL-1β in patients with EH in Xinjiang Kazakh [(61.0±4.1) vs (43.0± 3.1) ng/L, P= 0. 002]. Both the mRNA and protein expression of NLRP3, Caspase-1 and IL-1β were ex- pressed in the peripheral blood T lymphocytes. Compared to the controls, the mRNA levels of NLRP3, Caspase-1 and IL-1β were significantly increased in patients with E H in Xinjiang Kazakh (0. 0018 ± 0. 0005 vs 0. 0005±0. 0001, P=0. 010, 0. 0205±0. 0062 vs 0. 0065±0. 0021, P=0. 024, 0. 0092±0. 0019 vs 0. 0033±0. 0011, P=0. 008, re- spectively) ; and the protein expressions of NLRP3, Caspase-1 and IL-1β were also up-regulated markedly. ConclusionThe peripheral blood T lymphocytes express endogenous NLRP3 inflammasome signaling pathway related molecules, and expressions of NLRP3 and its downstream factors Caspase-1, IL-1β are increased in patients with EH in Xinjiang Kazakh.
