摘  要:目的研究巴马小型猪T1DM模型血清25-羟维生素D3[25(OH)D3]变化,探讨巴马小型猪T1DM空腹血糖(FBG)与血清25(OH)D3的相关性。方法选用健康雄性巴马小型猪12只,分为对照组6只和糖尿病模型组6只。糖尿病模型组腹腔注射链脲佐菌素(STZ),第一次腹腔注射STZ 75 mg/kg,1周后第二次腹腔注射STZ 150 mg/kg,对照组注射等量柠檬酸钠溶液;实验结束后行葡萄糖耐量试验(OGTT)、胰岛素释放实验(IRT)及检测空腹25(OH)D3水平,并行胰腺组织病理学检查、胰岛β细胞免疫组化染色分析。结果 (1)自链脲佐菌素给药后13 d开始,模型组空腹血糖明显升高并稳定维持在7.6~17.1 mmol/L水平。(2)造模成功1周后,模型组OGTT各时点血糖水平均高于正常对照组(P〈0.01),IRT各时点胰岛素水平均低于正常对照组(P〈0.01)。(3)胰岛β细胞免疫组化结果提示,模型组胰岛中胰岛素染色阳性面积[(10.68±2.78)%]显著低于对照组[(43.63±2.83)%](P〈0.01)。(4)模型组血清25(OH)D3[(29.67±4.41)ng/ml]低于对照组[(35.83±+3.25)ng/ml](P〈0.05)。(5)模型组25(OH)D3水平与FBG成负相关(r=-0.338,P〈0.05)。结论 (1)采用腹腔多次注射STZ可以成功构建巴马小型猪1型糖尿病模型。(2)模型组小型猪血清25(OH)D3水平降低与T1DM的发生有关。Objective To study the changes of serum 25-hydroxyl vitamin D3(25(OH)D3) in Bama miniature pig model of Type 1Diabetes Mellitus(T1DM), and explore the correlation of Fasting Blood Glucose(FBG) of Bama miniature pig model of T1 DM and serum 25(OH)D3. Methods 12 healthy male Bama miniature pigs were randomly divided into a control group and a diabetic group,with 6 cases in each group. Diabetic model group was injected with streptozotocin(STZ), and 75mg/kg STZ was injected from abdominal and 150mg/kg STZ was injected from abdominal after one week. Furthermore, the same amount of Sodium Citrate Solution was injected in the control group. After the experiment, we did oral glucose tolerance test(OGTT), insulin release test(IRT)and fasting plasma 25(OH)D3level detection, pathological examination for pancreatic tissue and Immunohistochemical staining analysis for pancreas islet β cell. Results(1) After 13 days of STZ injecting, FPG in model group was elevated and maintained at the level of 7.6-17.1mmol/L.(2) After one week of successful modeling, glucose levels of each time point in model group were higher than that of normal control group(P0.01), IRT insulin levels of each time point were lower than the normal control group(P0.01).(3) The immunohistochemical results of Islet β cell suggested that the areas of positive staining of insulin islet in the model group[(10.68 2.78)%] were significant lower than that of the control group [(43.63 2.83)%](P0.01).(4) The levels of serum 25(OH)D3of model group [(29.67 4.41) ng/m L] were significant lower than that of the control group [(35.83 +3.25) ng/m L](P0.05).(5) The levels of 25(OH)D3in model group were negatively correlated with FBG(r=-0.338, P0.05). Conclusion(1) Repeated intraperitoneal injection of STZ can successfully build Bama miniature pig model of T1 DM.(2) The decrease of serum 25(OH)D3in miniature pig of model was associated with the occurrence of T1
