摘　　要：目的探讨胎盘特异性基因4(PLAC4)mRNA在不同孕期孕妇血浆中表达的动态变化。方法经羊水穿刺核型分析确诊为孕正常整倍体胎儿的单胎妊娠孕妇82例(对照组),按收集血样时的不同孕周范围(17-23周)进行分组,采用定量PCR法检测PLAC4mRNA的表达水平。同时对18例孕非整倍体胎儿孕妇血浆(病例组,其羊水核型分析均为21三体胎儿)PLAC4mRNA行定量分析。比较PLAC4mRNA在不同孕期及两组间的含量差异。结果对照组血浆中PLAC4mRNA在17-18周(中孕)时的表达水平低于20-23孕周(P<0.05);对照组与病例组间的胎儿PLAC4mRNA含量无统计学差异(P>0.05)。结论中孕期孕妇血浆中胎儿游离PLAC4mRNA含量在20-23孕周时较高;中孕期21三体孕妇血浆中胎儿基因PLAC4mRNA含量与孕正常整倍体胎儿的孕妇无统计学差异。Objective To explore the dynamic changes of plasma free fetus placenta-specific 4（PLAC4）gene mRNA concentrationin in pregnant women in different gestational weeks.Methods PLAC4 mRNA expression in plasma was detected by quantitative PCR in 82 single pregnant women with euploid pregnancies in different gestational weeks（17-23 gestational weeks,group C）and in 18 trisomy 21 pregnancies（20-25 gestational weeks,group T21）,who were all diagnosed by amniocentesis.PLAC4 mRNA expression levels were compared between two groups and among different gestation stages.Results PLAC4 mRNA expression level in group C was lower in pregnant women with 17-18 weeks of gestation than that in pregnant women with 20-23 weeks of gestation（P〈0.05）.PLAC4 mRNA expression level of group C was similar to that of group T21（P〉0.05）.Conclusion PLAC4 mRNA expression level is increased in the middle gestation,which has no significant difference between the pregnant women with euploid pregnancies and those with trisomy 21 pregnancies.
