摘 要:目的研究不加防腐剂尿液保存方法细菌繁殖情况及对24 h尿蛋白定量测定的影响。方法收集新鲜混合清晨尿液标本50份,分别用UF-1000i尿液有形成分分析仪和AU-640生化仪测定新鲜尿液、低温和常温保存24 h后尿液中细菌和微量总蛋白、微量白蛋白的变化。结果新鲜尿液低温保存24 h后,细菌数差异无统计学意义(P=0.520),而常温保存24 h后,细菌数增加了近13倍,与新鲜尿液相比细菌数差异具有统计学意义(P=0.000)。新鲜尿液低温及常温保存24 h后,微量总蛋白、微量白蛋白差异均无统计学意义(P>0.05),且保存前后微量总蛋白、微量白蛋白值的相关性均具有统计学意义(P=0.000)。结论低温和常温保存可分别抑制和促进细菌生长繁殖,但均不影响尿蛋白的测定值;常温保存尿液是24 h尿蛋白定量测定的安全、简便、可靠的方法。Objective To study the bacterium multiply in urine stored without antiseptics and its effect on quantitative assay of urinary protein in 24 hours. Methods Fifty fresh and mixed morning urine samples were collected,and the concentration of bacteria,micro-total protein and microalbumin in fresh urine and urine stored for 24 hours at low and ordinary temperature were detected with UF-1000 i urinary formed elements analyzer and AU-640 biochemical analyzer,respectively. The differences of every index between pre-and post-storage were analyzed with paired t-test. Results There was no statistically difference between the bacterium number in fresh urine and that in urine stored at low temperature( P = 0. 520),while the bacterium number in urine stored at ordinary temperature,which increased by about thirteen times,was statistically different from that in fresh urine( P = 0. 000). When compared with micro-total protein and microalbumin in fresh urine,neither of them in low and ordinary temperature-stored urine had statistically difference( P〉0. 05). Furthermore,the micro-total protein and microalbumin in low and ordinary temperature-stored urine were all statistically correlated with those in fresh urine( P =0. 000). Conclusion The low and ordinary temperature storage can inhibit and promote the growth and multiply of bacterium,respectively,which don't affect the value of urinary protein. The ordinary temperature storage is a safe,convenient and reliable method for urine collected for quantitative assay of urinary protein in 24 hours.
