摘  要:目的:探讨盐酸椒苯酮胺(piperphentonamine hydrochloride,PPTA)对豚鼠耳蜗缺血再灌注损伤(ischemia-reperfusion injury,I/RI)后凋亡细胞的保护。方法将24只豚鼠随机分为正常组、假手术组、I/RI组、PPTA组4组,每组6只。采用TUNEL法观察耳蜗细胞凋亡情况,计算凋亡指数(apoptotic index,AI),组间比较检测结果。结果正常组、假手术组和PPTA组耳蜗没有或仅个别部位有凋亡细胞,I/RI组凋亡细胞明显多于其它3组。结论细胞凋亡是耳蜗I/RI的一种损伤形式,PPTA可能通过减少细胞凋亡对I/RI耳蜗起保护作用。Objective To investigate the protection by piperphentonamine hydrochloride(PPTA) on apoptosis in cochlear of guinea pigs with Ischemia Reperfusion Injury(I/RI).Methods 24 healthy guinea pigs were randomly devided into the normal group,negative control group,I/RI group in which the cochlea of guinea pigs were apoptotic for I/RI, and PPTA group in which PPTA was injected into guinea pigs whose cochlea were with I/RI. Each group had 6 guinea pigs . The cell apoptosis of guinea pigs in every group was studied by TUNEL method.The difference between the groups was analyzed by apoptotic index (AI).Results No or few cell apoptosis was observed in cochlea in the normal group, control group and PPTA group . The cochlea apoptotic cells increased obviously in the I/RI group than the other three groups.PPTA could significantly reduce the apoptotic cells.Conclusion Apoptosis is a form of damage to the cochlea with I / RI.The therapy of PPTA may prevent the cochlea from I / RI by reducing apoptosis.
