摘 要:目的探讨Bcl-2/Bax表达与大鼠主动脉钙化的关系。方法 16只Sprague-Dawley大鼠随机分为对照组和钙化组,每组8只。钙化组大鼠给予维生素D3300 000 U·kg-1肌肉注射,每日1次;尼古丁25 mg·kg-1溶于花生油中早、晚各灌胃1次诱导大鼠血管钙化模型。对照组大鼠同时间点给予灌服等量生理盐水。干预6周后股动脉放血处死大鼠,Von Kossa染色观察主动脉形态变化,免疫组织化学法和免疫印迹法(Western blot)检测Bcl-2/Bax蛋白表达。结果钙化组大鼠主动脉血管中膜弹性纤维间可见大量黑色颗粒沉积,而对照组大鼠无黑色颗粒。免疫组织化学法检测结果显示,与对照组比较,钙化组大鼠主动脉平滑肌细胞中Bcl-2表达无明显变化(P>0.05),而Bax阳性表达率显著增高(P<0.05)。Western blot检测结果显示,与对照组比较,钙化组大鼠主动脉平滑肌细胞中Bcl-2蛋白表达无明显变化,Bax蛋白表达显著增加(P<0.05)。结论凋亡系统Bcl-2/Bax失衡可能为血管钙化的机制之一。Objective To explore the relationship between Bcl-2 / Bax and vascular calcification in rat aorta. Methods Sixteen Sprague-Dawley rats were randomly divided into control group and calcification group,with eight rats in each group.The rats in calcification group were received a single dose intramuscular injection of vitamin D3( 300 000 U·kg-1) in the morning and two doses of nicotine( 25 mg·kg-1dissolved in peanut oil) gavage in the morning and evening to induce vascular calcification. The rats in the control group were treated with the same amount of normal saline at the same time. After six weeks intervention,the rats were sacrificed by withdrawing blood from arteria cruralis,the morphologic changes of arteries were examined by Von Kossa staining. The expression of Bcl-2 / Bax was detected by immunohistochemistry and confirmed by Western blot. Results There were mass black granules deposited in elastic fibers of calcified rat aortic vascular media in calcification group,while it was not found in control group. The result of immunohistochemistry showed that the positive expression rate of Bax was more obviously in calcification group compared with control group( P〈0. 05),but there was no significant difference in the expression of Bcl-2 between the two groups( P〉0. 05). The result of Western blot showed that the expression of Bcl-2protein in smooth muscle cells in calcification group had no obvious change,while the expression of Bax protein increased more significantly compared with control group( P〈0. 05). Conclusion Imblance of Bcl-2 / Bax may be one of the mechanisms of vascular calcification.
