摘  要:目的对一起呼吸道感染疫情人腺病毒进行病原鉴定。方法将4株腺病毒核酸阳性患者标本接种于Hep-2细胞进行培养,利用产生病变的细胞提取核酸DNA,PCR扩增六邻体蛋白基因,对扩增的产物进行测序,并通过BLastn序列分析以确定其型别。结果 4株腺病毒毒株六邻体蛋白基因PCR产物测序和Blastn分析结果证明该腺病毒与人7型腺病毒同源性为最高,大于99%。结论本研究从患者标本中分离到的病毒毒株为腺病毒7型。Objective To isolation the adenovirus from samples taken from uationts with respiratory tract infection and to identify the type of the adenovirus. Method Four adenovirus positive sample from our were seeded onto Hep-2 cell.Extracted viral DNA from the CPE cell and Amplified the hexon gene via polymerase chain reaction( PCR). The PCR product was sequenced and the sequence was subjected alignment to the type 7 hexon gene with the tool BLASTn seaching in Gene Bank to identify the type of the adenovirus. Result The four adenovirus strain ware isolated. BLAST analysis on hexon gene showed that the adenovirus strain isolated was indentical to human adenovirus type 7 strain with more than99% homogeneity。Conclusion The positive human Adenovirus isolated from pations sample are identified as human adenovirus type7.
