摘  要:目的 研究瘢痕疙瘩成纤维细胞中人端粒酶逆转录酶和caspase-3的表达及二者间的关系,以及两种因子与瘢痕疙瘩发生、发展机制的关系.方法 以普通瘢痕为对照,采用RT-PCR和蛋白免疫印染法,研究瘢痕疙瘩成纤维细胞中,人端粒酶逆转录酶、caspase-3 mRNA和蛋白表达的变化,并进行比较分析.结果 瘢痕疙瘩成纤维细胞中,人端粒酶逆转录酶的mRNA和蛋白表达水平高于普通瘢痕(P<0.01);而活化caspase-3的表达水平活性显著低于普通瘢痕(P<0.05).结论 人端粒酶逆转录酶和caspase-3在瘢痕疙瘩成纤维细胞中的表达存在异常,它们对瘢痕疙瘩形成过程中的成纤维细胞异常增殖和凋亡起着重要的作用.Objective To study the expression and relationships of human telomerase reverse transcriptase and caspas-3 gene in keloid derived fibroblasts and the relationships between the expression of two genes and the occurrence & development mechanism of the keloid. Methods The normal sear as the control group, the chan- ges of human telomerase reverse transcriptase and caspase-3 expression were detected by semi-quantitative RT- PCR and Western blot methods, then the resuhs were analyzed. Results Compared with scar, The expression of mRNA and protein of human telomerase reverse transcriptase was higher in fibroblasts of keloid ( P 〈 0.01 ) , but the expression of activated caspase-3 was significant lower ( P 〈 0.05 ). Conclusion The expression of human telomerase reverse transeriptase and easpase-3 in fibroblasts of keloid is abnormal, they play important roles in the abnormal proliferation and apoptosis of fibroblasts in the course of keloid.
