摘 要:目的检测分析大麻性别连锁SCAR标记对于中国大麻性别的特异性,为植株性别鉴别研究积累资料。方法分别收集中国云南、新疆、甘肃、山西等4个不同地区大麻雌雄植株各15份,共计120份。参考文献设计合成SCAR标记引物,对样本进行扩增和琼脂糖凝胶电泳检测,并随机抽取样本进行盲测验证。结果 120份已知性别的大麻样本经检测,其中113份样本结果正确,总正确率94.2%;16份随机盲测样本经检测,15份结果正确,正确率为93.8%。结论 SCAR323与SCAR119标记可被用作中国大麻性别的鉴别。Objective To investigate sex-link, and SCAR mark specificity of Chinese Cannabis sativa, for the purpose to provide data and information for study and research of early sex identification of Chinese hemps. Methods 120 male and female hemp samples from four different regions of China (Yun Nan, Xin Jiang, Gan Su and Shan Xi), 15 male and 15 female respectively, were collected and tested by SCAR markers. SCAR marker design synthesis by reference, then tested through amplification and agarose gel electrophoresis, at the same time, it has a blind test certification by random samples. Results 113 of total 120 samples were correct, which reached an accuracy of 94. 2%. In a blind test of 16 samples of Cannabis sativa, the result of 15 samples complied with their sex phenotype. Conclusion SCAR323 and SCAR119 could be used for sex identification of Chinese hemp.
