摘  要:目的 研究Peramivir体外抗禽流感H7N9病毒效果 方法 采用体外抗病毒实验方法和流感病毒神经氨酸酶抑制实验并结合病毒序列分析的方法 结果 体外抗病毒实验发现三个抗病毒药物peramivir,oseltamivir和zanamivir针对禽流感病毒A/Anhui/1/2013(H7N9)的抗病毒有效浓度EC50分别是0.74±0.24 μmol/L,25.5±16.0μmol/L,29.4±6.2μmol/L,但是针对A/Shanghai/1/2013(H7N9)的有效浓度分别是74.4±18.7 μmol/L,>1000 μmol/L,和37.1±11.9 μmol/L,各自有100倍,大于40倍和1.2倍的升高.体外神经氨酸酶抑制实验发现三种药物对两个实验病毒均是敏感的.序列分析发现A/Shanghai/1/2013(H7N9)神经氨酸酶基因有R292K的突变,这种突变公认是oseltamivir耐药的表现.结论 在MDCK细胞上三种抗流感病毒药物均表现出对病毒A/Anhui/1/2013(H7N9)的有效性.但针对A/Shanghai/1/2013(H7 N9)病毒peramivir,oseltamivir有一定的抗性.判断一个病毒是否是耐药毒株需要结合神经氨酸酶抑制实验、序列分析和药物的抗病毒实验结果来综合分析.Objective To study the antiviral effects of peramivir against avian influenza A (H7N9) in vitro.Methods An antiviral assay in vitro and neuraminidase inhibition assay as well as sequencing were conducted to evaluate antiviral activity of peramivir against avian influenza A (H7N9) viruses.Results Effective concentration 50 (ECs0) of peramivir,oseltamivir and zanamivir against the A/ Anhui/1/2013 (H7N9) (abbreviated as Anhui/1) were 0.74 ± 0.24 μmol/L,25.5 ± 16.0 μmol/L,29.4 ± 6.2 μmol/L respectively,but EC5o of three neuraminidase inhibitors (NAIs) against A/Shanghai/1/2013 (H7N9) (abbreviated as Shanghai/1) were 74.4 ± 18.7 μmol/L,〉 1000 μmol/L,and 37.1 ± 11.9 μmol/L,with 100,〉 40 and 1.2 fold increasing compared with the EC50 against Anhui/1 respectively.The susceptibility of avian influenza (H7N9) viruses Anhui/1 and Shanghai/1 to NAIs showed that NAIs were sensitive to both viral strains.But the strain Shanghai/1 has a lysine at position 292 of the NA gene instead of arginine at this site.The mutation R292K was thus supported the resistance to oseltamivir.Conclusion Neuraminidase inhibitors have been shown to be effective for the treatment of A/Anhui/1/2013 (H7N9) on MDCK cells.The resistance of neuraminidase inhibitors against A/Shanghai/1/2013 (H7N9) was determined at MDCK cells.It matched the feature of NA gene of this virus.Comprehensive consideration should be taken when we indentify drug resistant virus.
