摘 要:目的观察汉防己甲素(Tetrandrine,TET)对小鼠脑缺血再灌注损伤的影响,从内质网应激(ERS)角度探讨其分子机制。方法随机将雄性Balb/c小鼠90只分为假手术组(S组,n=30)、缺血再灌注组(I/R组,n=30)与TET处理组(n=30),HE染色观察病理结果,神经功能缺陷评分评价脑损伤情况,2,3,5-氯化三苯基四氮唑(TTC)染色法检测脑梗死体积,干湿重法计算脑含水量,western blot法检测缺血侧大脑皮层ERS和凋亡蛋白表达。结果与S组比较,I/R和TET组神经功能学评分(NDS)评分和脑含水量升高(P<0.05),脑梗死体积增大(P<0.05),GRP78、CHOP和caspase-12表达上调(P<0.01);与I/R组比较,TET组NDS评分和脑含水量降低(P<0.05),脑梗死体积减小(P<0.05),GRP78、CHOP和caspase-12表达下调(P<0.05)。结论 TET显著减轻小鼠脑组织I/R损伤,与降低I/R诱导的ERS和ERS介导的细胞凋亡有关。Objective To investigate the protective effects of tetrandrine(TET) after mice ischemia reperfusion injury and analyze the molecular mechanism in the view of endoplasmic reticulum stress. Methods The male Balb/c mice were randomly divided into 3 groups: Sham group(S group, n = 30), Ischemia-reperfusion group(I/R group, n = 30),and tetrandrine group(TET group, n = 30). The pathological results were investigated by HE staining and the cerebral injury situation was evaluated by neurological deficit scores. Infract volume was measured by TTC staining, brain water content was measured by wet-dry weight method and expression of GRP78, CHOP and caspase-12 were measured by western blot. Results Compared with the S group, infarct volume, water content and expression of GRP78, CHOP and caspase-12 were significantly increased in I/R group and TET group(P〈0.05 or P〈0.01). Compared with the I/R group, infarct volume, water content, brain injury and expression of GRP78, CHOP and caspase-12 were significantly decreased in TET group(P〈0.05). Conclusion TET significantly alleviates focal cerebral ischemia-reperfusion injury in mice and relates to inhibition of the endoplasmic reticulum stress and apoptosis.
