摘 要:目的:HO-1基因对小鼠运动能力的影响及其抗运动性疲劳作用。方法:饲养、适应性运动BDF1小鼠,6周龄时随机分为生理盐水对照组、锌原卟啉药物抑制组、钴原卟啉药物诱导组、HO-1转基因组,每组均使用动物跑台进行力竭运动,并分为力竭后即刻组和力竭后24h组。利用RT-PCR、Western-blot、动力学法、两点法及考马斯亮兰法分别测定各组HO-1mRNA及蛋白表达量、血清CK、BUN、尿蛋白含量。结果:生理盐水组力竭运动后HO-1mRNA表达增加,其余三组变化不大,生理盐水组和药物抑制组力竭后即刻HO-1蛋白表达量明显高于运动前,运动后24h则明显下降,药物诱导组则力竭运动后24h HO-1蛋白表达量明显高于运动前和力竭运动后即刻水平,且运动前高于运动后即刻;药物诱导组和转基因组小鼠力竭运动时间较生理盐水组和药物抑制组长,且均有统计学差异;药物诱导组CK、BUN和尿蛋白清除均较快,药物抑制组则均较慢;转基因组CK、BUN清除较快,但尿蛋白始终处于较高水平。结论:力竭运动可以诱导野生型小鼠骨骼肌HO-1表达增加,对HO-1高表达小鼠影响不明显;一定范围内高表达HO-1能够增强小鼠运动能力,促进运动性疲劳的恢复。Objective : In order to study the H0 - 1 gene effects on exercise ability and resist- ance to fatigue in mice. Method: Breeding, adaptively exercise BDF1 mice, 6 weeks age are randomly divided into normal saline control group, zinc protoporphyrin pharmacological inhi- bition group, the cobalt protoporphyrin drug induced group, HO -1 genetically modified group, each group are used for exhaustion exercise on running machine, and divided into im- mediate group and 24h after exhaustion groups. HO - 1 expression, serum CK, BUN, urine protein are measured by using RT- PCR,Western -blot method, kinetic method, two point method and coomassie brilliant blue method. Results : HO - 1 mRNA expression of the saline group increases after exhaustion exercise, the other three groups changed little, HO - 1 pro- tein after exhaustion exercise of the saline group and drug inhibition group is higher than pre -exercise ,24h after exercise obviously declines, 24h after exhaustion exercise in drug in- duced group is higher than pre - exercise than immediately exercise. Exhaustion exercise time of the drug induced group and genetically modified mice is longer than pharmacological inhi- bition and the saline group, significant variations are found in them. CK, BUN and urineprotein removal are faster in drug induced group, the pharmacological inhibition of the group are slower. CK, BUN clear faster in genetically modified group, but urine protein is in a higher level. Conclusion : Exhaustion exercise can induce skeletal muscle HO - 1 expression of the wild type mice, the high HO - 1 expression is not obvious in mice. Expression of HO - 1 must be within limits, which can enhance mice exercise ability, promote the recovery of exercise fatigue.
