摘  要:研究山丹黄参多糖对四氯化碳(CCl4)致小鼠肝损伤的影响.对40只小鼠先灌胃山丹黄参多糖(12.5,25,37.5g·L-1)和等量的生理盐水,然后腹腔注射0.2%的CCl4橄榄油溶液,称量小鼠体重、肝重变化,解剖观察小鼠肝脏的形态变化,显微镜观察肝组织结构的变化,免疫组织化学法检测TNF-α在肝组织表达的变化.研究显示,CCl4组小鼠体重明显减轻(P<0.01),肝重明显增大(P<0.01),肝脏明显肿胀,肝表面粗糙,有大量脂滴附着,肝脏周围亦有脂状物堆积;肝组织结构不清,细胞炎性坏死,TNF-α蛋白阳性表达明显增强(P<0.01);山丹黄参多糖各组与CCl4组相比体重显著或极显著增加(P<0.05或P<0.01),肝重明显减小(P<0.05),肝脏肿胀明显减轻,肝形态正常,肝脏表面脂滴减少,肝组织结构清晰,TNF-α蛋白阳性表达明显减少(P<0.01).结果表明,四氯化碳对肝组织有明显的损伤效应,山丹黄参多糖具有减少脂质沉着、抑制肝脏肿胀、促进肝脏机能稳定、抑制细胞过度凋亡等作用,能有效减轻四氯化碳对肝组织的损伤.To investigate the effects of Shandan Sphallerocarpus gracilis polysaccharide(SGP)on the liver histolomorph structure and the expression of TNF-α protein in liver tissue injury induced by carbon tetrachloride(CCl4)in mice,40 mice are gavaged with 0.2mL SGP at different concentrations(12.5,25,37.5g·L-1)or physiological saline,and then inject intraperitoneally with doses of 0.2mL CCl4solution(0.2 % in olive oil).The body weight and liver weight are recorded,the morphologic change of the samples are observed,and the change in the expression of TNF-αin liver tissue are determined by immunohistochemical method.Results show the body weights of CCl4 group are significantly decreased,while the liver weights are increased significantly(P〈0.01)and swelled markedly,surface is rough and presenting bright red.There are clear white fat droplets,the liver is surrounded by some white fat deposits,positive expression of TNF-αprotein increases significantly in liver(P〈0.01).However,the body weights in the SGP groups are significantly increased(P〈0.01),but the liver weights are significantly decreased(P〈0.05),the liver swollen is significantly mitigated,surface is smooth,there is a little white fat droplets,no white fat deposits around the liver,positive expression of TNF-αprotein decreases significantly in liver(P〈0.01).All results indicate that CCl4 has an obvious damage effect on the liver tissue.SGP can reduce lipid composure,inhibit the swollen of the liver and excessive apoptosis of liver cell,promote liver function stability,which can reduce liver tissue damage induced by carbon tetrachloride effectively.
