摘  要:目的:探讨华蟾素对多发性骨髓瘤RPMI 8226细胞株增殖、凋亡的影响及作用机制。方法:分别采用四氮唑蓝(MTT)比色法、流式细胞仪检测华蟾素对多发性骨髓瘤RPMI 8226细胞增殖、凋亡的影响。用ELISA检测细胞上清液中白细胞介素-6(IL-6)及血管内皮生长因子(VEGF)水平。结果:1不同浓度华蟾素对RPMI 8226细胞的增殖均有抑制作用,随药物浓度的增加及作用时间的延长,药物的作用效果增强;2不同浓度华蟾素(0.0625、0.125、0.25、0.5μg/m L)作用于RPMI 8226细胞72h,细胞凋亡率分别为(14.92±1.59)%、(19.17±2.10)%、(19.43±2.63)%和(27.47±3.39)%,与对照组(2.98±0.67)%比较,差异有统计学意义(P<0.01)。0.5μg/m L浓度华蟾素凋亡率高于其于各组(P<0.01);3与对照组比较,不同浓度华蟾素作用于RPMI 8226细胞72h,细胞上清中IL-6及VEGF水平下降,其中高浓度华蟾素组(0.25及0.5μg/m L)的变化有显著性差异(P<0.01),但低浓度组(0.0625及0.125μg/m L)变化无统计学意义(P>0.05)。结论:华蟾素在体外具有抗多发性骨髓瘤的作用,并呈时间和剂量的依赖性,与其直接细胞毒作用及诱导凋亡有关,降低瘤细胞分泌IL-6及VEGF的水平可能是其作用机制之一。Objective: To investigate the proliferation-inhibiting and apoptosis-inducing effects of cinobufacini on human myeloma RPMI 8226 cells and its mechanisms. Methods: Cell proliferation was measured by methylthiazolyl tetrazolium(MTT)assay. The rates of cell apoptosis were evaluated by flow cytometry(FCM). The IL-6 and VEGF concentrations of cell supernatants were tested by ELISA. Results: 1Cinobufacini showed significant effect of inhibiting proliferation on RPMI8226 cells at various concentrations from 0. 0625 to 0. 5μg / m L in a time and dose-dependent manner(P〈0. 01) and it also showed significant effects on cell apoptosis rate(P〈0. 01) when cultured with cinobufacini at the concentrations for 72 h in a dose-dependent manner. The rate of cell apoptosis in 0. 5μg / m L cinobufacini group was significantly higher than that of the other groups. Compared with that in the control group,the IL-6 and VEGF concentrations were down-regulated after acted by 0. 25 and 0. 5 μg /m L cinobufacini(P〈0. 01) but the levels of IL-6 and VEGF in 0. 0625 and 0. 125 μg / m L cinobufacini groups had no significant change(P〉0. 05). Conclusion: Cinobufacini can inhibit the proliferation of RPMI 8226 cells in a time and dose-dependent manner,which may be relevant to cytotoxic effect and promoting apoptosis. Its mechanisms may be correlated with the levels IL-6 and VEGF secretion down-regulating.
