摘 要:从富含偶氮染料的环境中取样,通过在含有偶氮染料的富集培养基中不断驯化培养,构建了一组高效稳定的偶氮染料降解复合菌群。该菌群在添加600 mg/L直接黑染料的降解培养基中55℃静置培养9 h,其脱色率可达83.56%。为进一步提高该菌群对偶氮染料的脱色效果,通过响应面分析法对其降解培养基进行优化。首先由Plackett-Burman试验确定影响染料直接黑降解的3个主要因素为葡萄糖、蛋白胨以及Fe Cl3,随后通过中心组合试验和响应面分析确定最优的培养基配方为(g/L):葡萄糖11.48、蛋白胨5.1、Fe Cl30.12、牛肉膏4.5、K2HPO42.7、Na H2PO45.25、Mg SO40.4和Mn SO40.05。经优化以后,在含有0.6 g/L直接黑染料的培养基中55℃静置培养9 h,该复合菌群对偶氮染料直接黑的脱色率高达99.24%,展现出了较大的应用潜力。A microbial consortium with high effective and stable azo dye degradation ability was constructed by successive domestication in the enrichment medium containing azo dyes,where the inoculums were sampled from the environment filled with azo dyes. The decolorization ratio in this microbial consortium could reach83.56% when incubated in the degradation medium containing 600 mg / L of direct black at 55 ℃ for 9 h.In order to further increase its decolourization ratio to azo dyes,response surface methodology was utilized to optimize the degradation medium.Firstly,Plackett-Burman test was utilized to identify the most significant factors influencing the decolourization ratio.It was found that the three main factors affecting the dye decolorization rate were Glucose,Peptone and Fe Cl3. Subsequently,central composite design and response surface analysis were utilized to optimize the degradation medium.It was found that the optimum combination( g / L) was: Glucose 11.48,Peptone5.1,Fe Cl30.12,Beef Extract 4.5,K2HPO42.7,Na H2PO45.25,Mg SO40.4 and Mn SO40.05. After optimization,the decolorization ratio of microbial consortium increased to 99.24% showing obvious application potential.
