匹伐他汀对大鼠心肌缺血再灌注损伤的保护作用

摘　　要：目的:探讨匹伐他汀对大鼠急性心肌缺血再灌注损伤的保护作用,为他汀类药物在缺血再灌注心肌的临床应用上提供理论依据。方法:成年雄性SD大鼠42只随机分为假手术组(Sham组)、对照组(IR组)及实验组(Statin组),每组14只。Sham组与IR组术前给予生理盐水2m L/d灌胃4周,Statin组给予匹伐他汀0.42mg·kg-1·d-1,溶解成2m L灌胃4周。制作大鼠心肌缺血-再灌注模型时假手术组冠状动脉下只穿线,不结扎,无缺血-再灌注发生。IR组、Statin组均结扎冠状动脉前降支(LAD)30min,再松解120min。术后检测血清中肌酸激酶同工酶(CK-MB)水平,用氯化三苯基四氮唑(TTC)及伊文氏蓝(Evans blue)双染色法测定各实验组心肌梗死面积及缺血区面积。结果:与Sham组相比,IR组、Statin组的血清CK-MB水平显著升高(P<0.01);Statin组CK-MB升高水平低于IR组(P<0.05)。I/R组、Statin组缺血区面积差别无统计学意义(P>0.05)。与I/R组相比,Statin组梗死面积显著减小(P<0.05)。结论:匹伐他汀可以减少缺血再灌注后心肌梗死面积,从而减轻心肌缺血再灌注损伤。Objective： To explore the positive role of pitavastain in a rat model of myocardial ischemiareperfusion,provide a theoretical basis for pitavastain clinical application in myocardial ischemia- reperfusion. Method： A total of 42 male SD rats were randomly assigned to three groups： Sham group,control group（ IR group） and experimental group（ Statin group）. In Sham group and control group,2m L saline were irrigated into stomach daily for 4 weeks,0.42 mg / kg pitavastain dissolved in 2m L saline were given in experimental group. In surgery,we weared line under the coronary artery without ligation in Sham group,and ligated the line for 30 minutes and then released it for 120 minutes in IR group and Statin group. The levels of CK-MB were discerned after operation. Triphenyl tetrazolium chloride and Evans blue double staining method was used to determinat the myocardial infarct size and ischemic area in each experimental group. Result：The levels of CK- MB in control group and experimental group elevated significantly compared with Sham group（ P〈0.01）. The level of CK- MB in IR group was significantly higher than Statin group（ P〈0. 05）.There was no significant difference in I / R and Statin group compared with ischemic area（ P〉0. 05）. But when it comes to infarct size,it was more smaller in Statin group（ P〈0.05）. Conclusion： Pitavastatin could reduce the cardiac infarct area and alleviate myocardial ischemia-reperfusion injury.
