摘 要:目的探讨活血通脉颗粒干预兔颈动脉粥样硬化(CAS)形成的作用机制。方法 75只新西兰白兔,随机抽取10只为正常组,其余65只采用高脂饲料喂养建立CAS模型。造模成功后将其随机分为模型组、自然消退组、活血通脉颗粒低、中、高剂量组、辛伐他汀组6组,每组7只。分别于实验第11周、第16周取颈动脉组织行HE染色以观察病理变化;ELISA法检测细胞间黏附分子-1(ICAM-1)、白介素-6(IL-6)表达水平;荧光定量PCR检测ICAM-1 m RNA表达。结果与自然消退组比较,活血通脉颗粒中、高剂量组和辛伐他汀组ICAM-1、IL-6水平明显下降(P<0.05),与活血通脉颗粒低剂量组比较,活血通脉颗粒高剂量组、辛伐他汀组明显下降(P<0.05);与自然消退组比较,各治疗组ICAM-1 m RNA表达明显下降(P<0.05)。结论活血通脉颗粒可能通过抑制ICAM-1、IL-6表达水平从而干预家兔动脉粥样硬化的形成。Objective: To invigorate the mechanism of Huoxue Tongmai Granule intervention the rabbit with carotid atherosclerosis(CAS).Methods: In the 75 New Zealand white rabbit,randomly selected 10 for the normal group,the remaining 65 rabbits were established by the high-fat feed carotid atherosclerosis model.After 11 weeks,the models were randomly divided into the model group,the natural healing group,the Huoxue Tongmai Granule low,medium and high dose group,simvastatin group with 7 rabbits in each group.11 weeks experiment respectively,and week 16 carotid line organization HE staining was used to observe the carotid artery tissue pathological changes.Meanwhile,ELISA method for detection of ICAM- 1 and IL-6 expression were dectected by the ELISA method.ICAM-1 m RNA expression was observed by the fluorescence quantitative PCR detection.Results: Compared with the natural healing group,ICAM-1 and IL-6 expression was decreased obviously in the Huoxue Tongmai Granule middle and high dose group(P〈0.05).Compared with Huoxue Tongmai Granule lowdose group,the high-dose group and the simvastatin group were significantly decreased(P〈0.05).Compared with the natural healing group,the m RNA expression of ICAM- 1 was significantly decreased in the treatment group(P〈0.05).Conclusion: Huoxue Tongmai Granule could inhibit ICAM-1and IL-6 expression level for the intervention of the formation of atherosclerosis.
