补中益气丸对糖尿病大鼠胃黏膜诱导型一氧化氮合酶及环氧化酶2表达的影响

摘　　要：目的观察补中益气丸对糖尿病大鼠胃黏膜诱导型一氧化氮合酶(i NOS)和环氧化酶2(COX-2)表达的影响,分析补中益气丸胃黏膜保护的作用机制。方法腹腔注射链脲佐菌素(STZ)结合30%乙醇灌胃建立糖尿病大鼠胃黏膜损伤模型,第5周起开始给药连续12周,观察大鼠体重及血糖变化,采用RT-PCR法和western-blot检测胃组织COX-2、i NOS的表达。结果大鼠持续消瘦且血糖居高不下,糖尿病成模率为75%。模型组COX-2mRNA、i NOSmRNA和中药组i NOSmRNA值均大于正常组(P<0.01),而中药组COX-2mRNA值小于模型组(P<0.05)。Western blot结果显示,模型组COX-2蛋白表达量大于正常组(P<0.05),中药组COX-2与正常组差异不显著(P>0.05),模型组和中药组i NOS均大于正常组(P<0.05)。结论糖尿病大鼠胃黏膜损伤严重,补中益气丸能调节COX-2和i NOS的表达,阻止胃黏膜损伤和糖尿病的进一步发展。Objective To observe the effects of Buzhong yiqi pills on the expression of iNOS and COX-2 in gastric tissue of diabetic rats, and to analyze the gastric mucosa protection mechanism of Buzhong yiqi pills. Methods The diabetic rat gastric mucosa damage model was established by intraperitoneal injection of streptozotocin （STZ） combined with orally administered ethanol. At the fifth week, drugs were given for 12 weeks, and body weight and blood sugar changes of rats were observed and the expressions of iNOS and COX-2 in gastric tissue were detected by RT-PCR and western blot. Results Rats pine away in succession and their blood sugar was staying at a high level, and the success rate of diabetes model was 75 %. Absorbance ratio of COX-2mRNA and iNOSmRNA in model group and TCM group were higher than that in normal group （ P 〈 0. 01 ）, COX-2mRNA absorbance ratio in TCM group is lower than that of model group （ P 〈 0.05）. The result of western blot analysis showed that COX-2 protein expression levels in the model group COX-2 were higher than that in the normal group （ P 〈 0.05）, but COX-2 in TCM group had no significant differences with normal group （ P 〉 0.05）. iNOS in model group and TCM group were significant higher than that in normal group （ P 〈 0.05）. Conclusion Gastric mucosa of diabetic rats was seriously damaged, buzhong yiqi pills can regulate the expression of iNOS and COX-2, preventing development of gastric mucosal lesion and diabetes.
