摘 要:目的:探讨红景天苷(salidroside,SAL)对大鼠离体肠系膜动脉的舒缩作用及调控机制。方法:应用DMT微血管张力测定法和ELISA法研究红景天苷对血管的影响及机制。结果:SAL对KCl预处理的血管环起收缩作用;对PE预处理的血管起舒张作用,其中内皮完整血管比去内皮血管舒张明显;L-NAME抑制SAL舒血管,亚甲蓝、吲哚美辛对SAL舒血管无影响;SAL升高血管NO和NOS。结论:SAL对血管具有收缩和舒张双重作用,缩血管可能与电压依赖性钙通道有关;舒血管可能与抑制内钙释放有关,具有与NO释放和NOS合成有关的内皮依赖性,但与环氧合酶途径无关。Objective: To observe vasomotor effect of salidroside( SAL) on rat mesenteric artery,and to explore its underlying regulatory mechanisms. Method: Tension was measured by DMT system to evaluate the vasomotor effect of SAL on rat endothelium-intact and endothelium-denuded mesenteric aortic rings. Different antagonists and ELISA method were used to illustrate the mechanisms of vasomotor effect of SAL. Result: 1 SAL had significantly vasoconstrictive effect on mesenteric aortic rings precontracted by KCl. 2 SAL relaxed endothelium-intact and endothelium-denuded mesenteric artery precontracted by PE in concentration-dependent manner,while the vasodilation of endothelium-intact mesenteric artery was significantly stronger than that of endothelium-denuded one. 3L-NAME attenuated the vasodilation of endothelium-intact mesenteric artery induced by SAL,while neither indomethacin nor methylene blue affected it. 4 Using ELISA,SAL can significantly increase NO and NOS of mesenteric artery. Conclusion: The results indicate that SAL plays a dual role of vasoconstriction and vasodilation in regulating mesenteric artery. Vasoconstriction may be associated with voltage-dependent calcium channel,while vasodilation may be associated with inhibition of intracellular calcium release. Besides,vasodilation shows endothelium dependence,which is related to NO released and NOS synthesized,regardless of the COX pathway.
