摘  要:目的:探讨中期因子(midkine,MDK)在食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)中的表达及其生物学功能。方法:收集本院胸外科行手术切除的食管鳞状细胞癌组织及对应癌旁组织60例,采用免疫组化染色方法检测肿瘤组织及对应癌旁组织中MDK的表达情况,统计分析MDK表达与肿瘤临床病理特征间的相关性;体外培养人食管鳞状细胞癌Eca-109细胞,利用siRNA特异性敲低细胞内MDK的表达水平,分别采用MTT及流式细胞仪检测Eca-109细胞增殖及凋亡能力的变化。结果:与癌旁组织相比,ESCC组织中MDK呈明显高表达趋势(P<0.05);ESCC组织中MDK高表达与肿瘤体积增大(>3cm)、肿瘤淋巴结转移及高TNM分期(III+IV期)相关(P<0.05);在体外,敲低MDK表达可显著抑制Eca-109细胞增殖能力,并促进细胞凋亡的发生(P<0.05)。结论:中期因子与食管癌组织生长有关,特异性下调食管癌细胞内MDK的表达能够显著抑制食管癌细胞增殖并促进其凋亡的发生。Objective; To investigate the expression and biological functions of midkine (MDK) in human esophageal squamous cell carcinoma (ESCC). Methods A total of 60 ESCC tissues and matched tumor adjacent tissues were collected. The expression of MDK in ESCC and matched tumor adjacent tissues were detected. The relationship between the expression of MDK and clinieopathlogical features was analyzed by Pearson Chi-square test. siRNA was used to down-regulate the expression of MDK in ESCC cells. The effect of MDK for cell proliferation and apoptosis were measured by MTT assay and FCW assay, respectively. Results The expression level of MDK was significantly higher in ESCC tissues than in tumor adjacent tissues (P〈 0.05). The high expression of MDK protein was positively correlated with large tumor size (〉3 cm), lymphatic metastasis and advanced TNM stage (Ⅲ+ Ⅳ) (P〈0.05). Down-regulation of MDK in ESCC cells could significantly suppressed cell proliferation and enhanced apoptosis. Conclusions MDK is overexpressed in esophageal squamous cell carcinoma tissues and as-sociated with tumor malignant clinicopathological features. MDK may promote ESCC progression by inducing cell growth.
