摘  要:目的优化苦参方的乙醇提取工艺。方法以乙醇浓度、溶剂倍量、提取时间为自变量,以K(苦参碱与氧化苦参碱总和)提取率及干膏得率的总评"归一值"为因变量,通过对自变量各水平进行多元线性回归及二项式拟合,选择合适的拟合方程模型,采用效应面法优选最佳工艺,并进行预测分析。结果最优提取工艺条件为8倍量65%乙醇,提取2次,每次2.5 h,总评"归一值"实测值与预测性偏差为-2.26%,二项式拟合复相关系数R2=0.949 9。结论星点设计-效应面法优选的提取工艺稳定可行,预测性良好。Objective To optimize alcohol extraction technology of Kushen recipe. Methods Independent variables were ethanol concentration,solvent amount and extraction time,dependent variable were the "Overall Desirability" of extraction rate of K(the sum of matrine and oxymatrine) and the yield of extract. Linear or nolinear mathematic models were used to estimate relationship between independent and dependent variables.Select the appropriate fi tted equation model, response surface method was used to optimize extraction,prediction was carried out through comparing observed and predicted values. Results Optimal extraction technology was as following: extracted 2 times with 8 times the amount of 65 % ethanol,2.5 h each time.The bias of the "Overall Desirability" between observed and predicted value was-2.26 %.Regression coefficients of binomial fitting complex model was as high as 0.949 9. Conclusion Optimized extraction technology was stable and feasible with high predictability.
