摘 要:莱氏野村菌微菌核已成功实现了液体诱导培养和对其分子发育机理的研究。为实现微菌核的规模化生产,我们通过对不同类型的接种体、不同培养时间的接种体和不同接种量对莱氏野村菌微菌核产量和生物量进行了研究。结果显示,培养24-48 h的菌体能够作为液体一步发酵的有效接种体,微菌核产量均达到1.0×10^4微菌核/mL。并优化莱氏野村菌微菌核液体30 L发酵工艺,产率达到20%-30%。研究结果为莱氏野村菌微菌核的规模化生产提供了科学数据参考。Microsclerotia (MS) formation was successfully induced in Nomuraea rileyi (Ascomycetes:Hypocreales) in liquid culture and the molecular mechanism of MS formation had investigated through comparative transcriptome analysis. To determinate the large scale fermentation, the various inoculums, the different culture times of mycelium and conidium inoculum, and the various inoculum size, affecting N. rileyi MS yields and biomass, were investigated. The results showed that N. rileyi MS yields were 1.0×10^4 MS/mL with inoculums, cultured in 24-48 hours at 25℃and the mycelia could be used as the effective inoculums for one step fermentation. Furthermore, N. rileyi MS yields of fermentation in 30 liters bioreactor were optimized and up to 20%-30%. The results will provide scientific data for N. rileyi MS fermentation in large scale.
